Abstract:The object of this study was to discover an alternative therapeutic agent with fewer side effects against acne vulgaris, one of the most common skin diseases. Acne vulgaris is often associated with acne-related bacteria such as Propionibacterium acnes, Staphylococcus epidermidis, Staphylococcus aureus, and Pseudomonas aeruginosa. Some of these bacteria exhibit a resistance against commercial antibiotics that have been used in the treatment of acne vulgaris (tetracycline, erythromycin, and lincomycin). In the current study, we tested in vitro antibacterial effect of chitosan-phytochemical conjugates on acne-related bacteria. Three chitosan-phytochemical conjugates used in this study exhibited stronger antibacterial activity than that of chitosan (unmodified control). Chitosan-caffeic acid conjugate (CCA) showed the highest antibacterial effect on acne-related bacteria along with minimum inhibitory concentration (MIC; 8 to 256 µg/mL). Additionally, the MIC values of antibiotics against antibiotic-resistant P. acnes and P. aeruginosa strains were dramatically reduced in combination with CCA, suggesting that CCA would restore the antibacterial activity of the antibiotics. The analysis of fractional inhibitory concentration (FIC) indices clearly revealed a synergistic antibacterial effect of CCA with antibiotics. Thus, the median sum of FIC (∑FIC) values against the antibiotic-resistant bacterial strains ranged from 0.375 to 0.533 in the combination mode of CCA and antibiotics. The results of the present study suggested a potential possibility of chitosan-phytochemical conjugates in the control of infections related to acne vulgaris.

The integration of traditional and modern medicine has gained increased recognition globally. In light of vast potentiality of medicinal plants as therapeutics, the present endeavor deals with the assessment of antifungal and synergistic activity of Urginea indica (Kunth.) (Family: Liliaceae) commonly known as wild onion against human pathogenic fungi viz., Aspergillu niger (MTCC 872) and Candida albicans (MTCC 183) procured from IMTECH, Chandigarh, India. The antifungal and synergistic activity was assessed by agar well diffusion method. The results were analyzed statistically using ANOVA with DMRT. The qualitative phytochemical analysis, column chromatography, TLC, MIC, and MFC was performed following standard protocols. Finally, the purified fraction was chemically characterized by HPLC, UV-Vis, FT-IR, (1 H & 13 C) NMR, and ESI-MS. The antifungal activity profile showed that acetone root extract exhibited broad-spectrum antifungal activity. However, in case of C. albicans, the extract exhibited significantly higher activity index. The qualitative analysis revealed the presence of varied phytochemicals. The extract exhibiting the highest activity was purified and tested for its synergistic or antagonistic potentiality against fungi revealed that the bioactive compound exhibited statistically significant synergism with three different commercially available antifungals viz., clotrimazole, ketoconazole, and fluconazole. Finally, the purified fraction was chemically characterized. The combination therapy against human pathogenic fungi seems to be a boon for patients with severe infections caused by multidrug-resistant pathogens. Thus, in current scenario, there is an urgent need to target new dimensions towards alternative antifungal therapy to preserve better human health for generations ahead.

Fungal infection mostly caused by marine oomycetes had hindered crustacean production thus searching for natural and safe treatment is currently needed. Thus, this study was conducted to investigate the antimycotic effect of different seaweed extract against marine oomycetes Lagenidium spp. and Haliphthoros spp. Two seaweeds species Eucheuma cottonii and Caulerpa lentillifera were extracted using ethanol, methanol and water. Each extracts was tested on four fungi strains of marine oomycetes species for minimum inhibitory concentration MIC and fungicidal activities. C. lentillifera ethanol extract showed the highest antifungal effect where it can inhibit three from four fungal strains. Meanwhile, E. cottonii ethanol extract has lowest MIC 500 ppm and inhibit L. thermophilum IPMB 1401 and H. sabahensis IPMB 1402 hyphal growths. Antimycotic effect on zoospores production shows reduction in production after 12 h immersion for three marine oomycetes species. Seaweed extracts toxicity on Artemia sp. showed approximately 5% mortality at 12 h immersion. It is suggested that 12 h immersion of seaweed extract is a suitable treatment for marine oomycetes in aquaculture. This study does not only show potential alternative control method for crab larvae health management, it may also contribute to the sustainable development and food security of aquaculture industry.
