Antifatigue effects of troxerutin on exercise endurance capacity, oxidative stress and matrix metalloproteinase‐9 levels in trained male rats

Abstract: The aim of this study was to investigate effects of troxerutin (TRX) on endurance capacity, oxidative stress and matrix metalloproteinase-9 (MMP-9) levels in trained male rats. Forty male Wistar rats were divided into five groups. The control (Vehicle) and exercise training (5 days/week) with vehicle treatment (Exercise), exercise training with TRX treatment at 75 (Ex-TRX75), 150 (Ex-TRX150), and 300 mg/kg (Ex-TRX300). The treated groups received TRX by gavage every day while the other groups received water fo… Show more

“…Importantly, growing evidence suggests that overproduced free radicals can damage proteins, lipids and DNA in the contracting myocytes, mediate activation of calpains and caspases, and modulate cellular signaling pathways including the mitogenactivated protein kinase (MAP kinase) and nuclear factor κ-B (NFκB) pathways [27,32]. The superoxide dismutase (SOD) and glutathione peroxidase (GPX) are the principal endogenous antioxidant enzymes to attenuate the damage induced by excessive oxidative stress, which by scavenging free radicals and their metabolites [8,27,45]. Malondialdehyde (MDA) is one of the degradation products of membrane lipid peroxidation caused by free radicals, which is an important indicator for evaluating cellular oxidative stress [8,33].…”

“…SOD is involved in the scavenging of oxygen free radicals in the body and thereby prevents lipid peroxidation and protects the cell membrane from oxidative damaged [ 24 ]. MDA is one of the products of lipid peroxidation induced upon injury and can aggravate the damage to the cell membrane.…”

“…A weighting (steel ring) equivalent to 5% of body weight attached to the tail root of each mouse. The animal exhaustion time was recorded when they failed to come up to the water surface for breathing within 5 s. Animals were then removed from the pool, dried with a paper towel and returned to their cages . To control the effect of circadian rhythm, all tests were performed between 07:00 and 14:00 in the order of EPM, vertical grid holding test, and forced swimming test (FST), respectively.…”
