Dipeptidyl peptidase-4 (DPP-4) is a proteolytic enzyme responsible for the rapid degradation of Glucagon-like peptide 1 (GLP-1) that is required for the secretion of insulin. DPP-4 also influences activation of node like receptor family, pyrin domain containing 3 (NLRP3) inflammasome under diabetic conditions. Although several polyphenols are reported for various bioactivities, they are consumed as part of the food matrix and not in isolation. Horsegram (Macrotyloma uniflorum) is a rich source of myricetin (Myr) (35 μg/g flour), reported for its anti-hyperglycemic effect. In this investigation, we aimed to study the effect of Myr, singly, and in the presence of co-nutrient horsegram protein (HP) on DPP-4 activity and its consequential impact on GLP-1, insulin, and NLRP3 inflammasome in high-fat diet and single low dose streptozotocin (STZ)-induced diabetic male Wistar rats. In diabetic control (DC), the activity of DPP-4 and its expression were higher compared to treated groups. The consequential decrease in the circulating GLP-1 levels in the DC group, but not treated groups, further indicated the effectiveness of our test molecules under diabetic conditions. Specifically, Myr decreased DPP-4 activity and its expression levels with enhanced circulating GLP-1 and insulin levels. Myr administration also resulted in a lessening of diabetes-induced NLRP3 inflammasome activation and enhanced antioxidant enzyme activities. HP also proved to be efficient in reducing elevated blood glucose levels and enhancing antioxidant enzyme activities. However, Myr, in the presence of HP as a co-nutrient, had diminished capacity to inhibit DPP-4 and, consequently, reduced potential in ameliorating diabetic conditions. Myr proved to be a potent inhibitor of DPP-4 in vitro and in vivo, resulting in enhanced circulating GLP-1 and insulin levels, thereby improving diabetic conditions. Though Myr and HP, individually ameliorate diabetic conditions, their dietary combination had reduced efficiency.

Acid phosphatases play a crucial role in food processing industries to reduce phosphate content of food. Here in acid phosphatase from the seeds of has been purified to homogeneity using UNOsphere-S cation exchange chromatography followed by gel filtration with 81.85 fold purification. Molecular weight of purified enzyme was 55,000 (± 1040) Daltons under physiological conditions. It was a heterodimer of subunits having molecular weights 27,093 and 28,241 Daltons as determined by MALDI-TOF analysis. The optimum pH and temperature for the purified enzyme was 5.0 and 50 °C respectively. The enzyme was stable in the pH range 3.5-5.5 and showed temperature stability up to 60 °C. Substrate specificity of enzyme was checked with differentsubstrates namely, p-nitrophenyl phosphate (p-NPP), ATP, ADP, glucose 6-phosphate, glucose-1-phosphate, fructose 6-phosphate, phenyl phosphate, α-naphthyl-phosphate, pyridoxyl phosphate and β-glycerophosphate. Enzyme showed high substrate specificity towards p-NPP, phenyl phosphate, ATP and α-naphthyl phosphate. K and V of enzyme were found to be 0.934 mM and 1.333 mM/min respectively with respect to p-NPP as a substrate. Chemical modification studies showed that tryptophan was present at the active site of the enzyme.

Macrotyloma uniflorum (Lam.) Verdc commonly known as horse gram is a kind of legume of tropics and subtropics. Its medicinal uses are known to Ayurveda and Sri Lankan traditional physicians for centuries. Present study was undertaken to collect data on medicinal uses and nutritional values of M. uniflorum. Ayurveda pharmacodynamic properties of M. uniflorum are Kashaya Rasa, Laghu, Ruksha, Tikshna Guna, Ushna Veerya and Katu Vipaka. Various medicinal preparations such as Dhanyamla and decoctions are prepared using seeds of M. uniflorum. It is mainly used as a tonic, astringent, diuretic and also recommended in rheumatism, neuralgia and other several diseases. Horse gram seeds are rich in natural phenols; mostly phenolic acids, flavonoids and the major anti-oxidants. Horse gram is considered as animal fodder and its full potential as a part of human diet has not been exploited completely. It can be consumed as seeds, as sprouts or as meal by itself. Horse gram is an excellent source of protein (22-24%). Seeds contain carbohydrates (57.2%), fat (1.1%), vitamins, minerals (3.2%) and good amount of soluble fibers. Extract of these seeds shows potent anti-adipogenic, anti-hyperglycemic anti-hyper cholesterolemic activities. It acts against oxidative stress. It is concluded that Horse gram can be used as a multifaceted treatment as well as a wholesome food.
