Problem statement: Poor glycemic control and oxidative stress is implicated as a common pathway in the development of diabetic neuropathy. Approach: In the present study, we investigated the protective effects of Enicostemma littorale Blume (EL) (2.5 g kg −1 ), a hypoglycemic and antioxidant herbal medicine in alloxan-induced diabetic neuropathy in male Charles foster rats.Results: Tail flick latency in hot immersion test and the formalin test were used to evaluate nociceptive responses in diabetic rats compared to non-diabetic rats. Preventive treatment of EL (for 45 days) and standard drug glibenclamide (Glib) significantly improved nociception in diabetic rats. The changes in lipid peroxidation status and anti-oxidant enzymes (super-oxide dismutase, glutathione peroxidase and catalase) levels observed in diabetic rats, were significantly restored by EL and glib treatment. Decrease in Na-K + ATPase activity was also significantly restored by EL. Glibenclamide was used as standard drug in present study. Conclusion: This study provided experimental evidence to preventive effect of EL on nerve function and oxidative stress in animal model of diabetic neuropathy. Hence EL may be tried clinically for the treatment of diabetic neuropathy since it was used as folk medicine in diabetic patients.

Problem statement:The Amino glycoside antibiotics cause drug induced nephrotoxicity at therapeutic doses and thus limits the use clinically. Oxidative stress is one of the main reasons for the development of nephrotoxicity. Approach: In light of this the aim of the present study was to investigate the role of mitochondrial as well as post-mitochondrial oxidative stress in the development of Gentamicin (GM) induced nephrotoxicity and to elucidate the role of Enicostemma Littoral blame (EL) extract, as a promising antioxidant therapy. Rats were divided into four groups, 1) (Control 2) GM (i.p., 80 mg kg −1 day −1 for 8 days) (treated 3) GM+EL treated (i.g, 2.5 gm kg −1 day −1 ) and (4) GM+ Vitamin C (VC) treated (i.g, 600 mg kg −1 day −1 ). GM treated animals showed high oxidative stress in mitochondrial as well as post-mitochondrial fractions of renal tissues as evidenced by increased lipid per oxidation levels, decrease in GSH content and activities of antioxidant enzymes, SOD and GPx. Results: Oxidative stress was more pronounced in mitochondrial fraction as compared to post-mitochondrial fraction. GM-induced nephrotoxicity was further corroborated by an increase in serum cretonne and Blood Urea Nitrogen (BUN) levels and altered kidney histopathological observations. Treatment with EL ameliorates antioxidant defense system of mitochondrial as well as post-mitochondrial fraction, with better improvement seen in mitochondrial fraction. Conclusion: The present study explored beneficial effect of EL extract as an antioxidant therapy to counteract mitochondrial and post-mitochondrial oxidative stress generated in kidney upon GM-treatment, thus prevented nephrotoxicity.

Background: Enicostemma axillare (Lam.) Raynal is used in traditional practice for the treatment of diabetes, malaria and liver disorders. No systematic toxicity study was described for this plant and hence the present was undertaken to evaluate acute and sub-acute toxicity of aqueous extract of Enicostemma axillare (AEEA). Objective: The acute oral toxicity study of AEEA was carried out as per the OECD guidelines 423 in mice and the sub-acute toxicity was carried out as per the guidelines set by OECD 407 in male and female rats. Materials and Methods: Body weight, food and water consumption, hematological parameters, biochemical parameters, organ weight and histopathological analysis were carried out. Results: No gross toxicity and mortality was observed upto a dose of 2000 mg/kg. For sub-acute toxicity test, 200 mg/kg and 400 mg/kg daily dose of AEEA administered orally for 28 days in male and female group of rats not exhibited any signs of toxicity and mortality.
