The present study focuses on in vitro antioxidant and enzyme inhibitory activity of three different solvent extracts (Methanol, Ethyl acetate and Hexane) of 3 different seaweeds viz: Sargassum wightii, Caulerpa racemosa, and Acanthophora spicifera. The preliminary phytochemical analyses of the seaweed extracts have revealed the presence of various phytochemicals. The antioxidant activities of the seaweed extracts have shown the scavenging effect. Among the extracts, SWEA, SWME and SWHE have exerted effective antioxidant activity with the IC 50 values (µg/mL) of: 32.86, 130.1 and 212.8, respectively. And similar trend of α-amylase and α-glucosidase activity/inhibitory property by seaweeds have been recorded. Hence, the ethyl acetate extract of S. wightii was subjected to gas chromatography. All the seaweed extracts were characterized through HPLC and FTIR analyses. The GC-MS analysis of ethyl acetate extract of S. wightii showed the presence of a bioactive compound, Heptadecanoic acid that might be have been the reason for the recorded inhibitory activity.

Evaluation of Saponin Extract from Vitex doniana and Pentaclethra macrophylla for Antibacterial Activity

Saponins are pharmacologically active compounds that have been shown to ameliorate abnormal physiological processes and be aptly applied in folklore for the treatment of maladies occasioned by infectious agents. Consequently, saponins from Vitex doniana and Pentaclethra macrophylla were evaluated for antibacterial properties, as these herbs are used in folk medicine. Dried pulverized plant materials were defatted, and solvents with varying polarity were applied at varying ratios for the extraction of saponins. Phyto-chemistry was in accordance with standard methods, while an antibacterial assay was made through the agar well diffusion and micro broth dilution techniques. Phytochemical quantitation showed high concentrations of tannins, 231˘0.6 CE/g, and saponins, 58% from V. doniana. Similarly, P. macrophylla stem bark extract also showed high concentrations of tannins, 309˘2.42 CE/g, alkaloids, 71%˘0.5%, and saponins, 87%˘3.4%. The ethanol extracts of V. doniana inhibited the growth of Staphylococcus aureus (ATCC 11775) and a clinical strain with inhibition zone ranges of 15.5˘2.12 to 7.0˘0.0 (mm) against leaf extracts and 20.0˘1.41 to 7.0˘0.0 (mm) against stem bark extracts. Conversely, saponin extract from V. doniana showed a broad spectrum of activity, as it inhibited both Gram-negative and -positive test strains, E. coli clinical strain (20.0˘1.41 mm), P. aeruginosa clinical strain (18.5˘0.71 mm), E. coli ATCC 11775 (17.0˘0 mm), and S. aureus clinical strain (13.0˘1.41 mm). However, a broad spectrum was similarly achieved with P. macrophylla extracts, as all test bacteria genus was susceptible. Saponin fractions showed a high potency and broad spectrum antibacterial activity and thus a validation of the folklore applications and the potential for use as a drug or drug scaffold.

Coreopsis tinctoria capitula (CTC) of the Compositae family has been used traditionally to treat various diseases in China, particularly type 2 diabetes mellitus (T2DM). This study evaluated the anti-lipid peroxidation, α-glucosidase and α-amylase inhibitory effects of CTC extracts, and analyzed its chemical composition by HPLC. Moreover, the antioxidant activity and protection effects of CTC extracts were investigated on high-fat/highsugar and streptozotocin-induced T2DM mice. In vitro study, the ethyl acetate extract (EAE) and butanol extract (BE) of CTC exhibited anti-lipid peroxidation (IC 50 : BHA > BE or EAE > ascorbic acid, p < 0.05) and α-glucosidase inhibitory activity (IC 50 : BE > EAE, p < 0.05). In vivo, the BE at the dose of 600 mg/kg was intragastrically given to T2DM mice, which exhibited a certain extent of repair and improvement of the levels of CAT, GSH, GSH-P X , SOD, as well as plasma biomarkers, compared with those in the model group (p < 0.05). These results demonstrated that CTC extracts have a positive effect to treat T2DM and it can be used for the treatment of T2DM in the future.
