Antidepressant-like effect of fluoxetine may depend on translocator protein activity and pretest session duration in forced swimming test in mice

Abstract: The antidepressant-like effect of fluoxetine (20 mg/kg i.p.) has been assessed using the forced swimming test (FST) in IRC (CD-1) mice exposed or not to a pretest session of different duration (5 or 20 min). The influence of the mitochondrial translocator protein (TSPO) activity on the antidepressant-like effect of fluoxetine (20 mg/kg i.p.) in the FST was also studied. The antidepressant-like effect of fluoxetine was observed only in mice subjected to a 5-min pretest session 24 h before the FST. The TSPO anta… Show more

“…Over the last three years, the FST has been used as the main test for evaluating both the modelling and drug-therapeutic efficiency. However, the main problem with the studied drugs, including fluoxetine, tipepidine, and tramadol, was the low statistical power for clinical applications, which may not provide a direction for meta-analyses [91,92,93].…”

“…Fluoxetine (100 mg) was combined with 40 ml of distilled water to prepare the drug solution, so there was 2.5 mg of the drug in each cc of solution [16]. After the preparation of these solutions, different doses were adjusted for each experimental group and orally gavaged daily at 9-12 A.M. for one week [17][18][19][20][21][22][23][24]. Animals received oral gavage of distilled water, herbal solution, and drug solution as follows: Group 1: distilled water gavage (the same volume as other groups) Group 2: 10 mg/kg of fluoxetine solution gavage Group 3: 10 mg/kg of herbal solution gavage Group 4: 5 mg/kg of herbal solution gavage Group 5: 2.5 mg/kg of herbal solution gavage…”
