Anticancer Profile of Rhodanines: Structure–Activity Relationship (SAR) and Molecular Targets—A Review

Abstract: The rhodanine core is a well-known privileged heterocycle in medicinal chemistry. The rhodanines, as subtypes of thiazolidin-4-ones, show a broad spectrum of biological activity, including anticancer properties. This review aims to analyze the anticancer features of the rhodanines described over the last decade in the scientific literature. The structure–activity relationship of rhodanine derivatives, as well as some of the molecular targets, were discussed. The information contained in this review could be of… Show more

“…To avoid these risks, it is crucial to seek new molecules with beneficial medical properties for targeting cancer cells. In 2021, nearly one-third of all drugs adopted for healthcare interventions were new anti-tumour agents, which were affirmed by the U.S. Food and Drug Administration [ 92 ]. In this context, thiazolidinone (heterocyclic compound) and its derivatives have been the centre of attention for many years.…”

“…However, epalrestat containing a rhodanine fragment was successfully used to treat diabetes mellitus [25]. For compounds containing fragments of rhodanine, antibacterial activity [26][27][28][29][30][31], fungicidal [26,32], antiviral [30], anti-tuberculosis [33], anti-inflammatory [26,27,34], and anti-cancer [26,27,30,[35][36][37] activities were noted. These compounds can be used in the treatment of diabetes mellitus [25,27], Alzheimer's disease [26,27], and a number of other diseases [38,39].…”

“…Such a group of derivatives may be compounds with a thiazolidin-4-one core. Thiazolidin-4-one derivatives exhibit a broad spectrum of therapeutic applications and hold clinical significance in the field of medicinal chemistry [ 9 , 10 , 11 , 12 , 13 ]. For example, thiazolidine-2,4-diones are a well-known group of antidiabetic drugs, glitazones, that act by activating PPARγ.…”
