Anticancer and antimicrobial properties of novel η6-p-cymene ruthenium(ii) complexes containing a N,S-type ligand, their structural and theoretical characterization

Abstract: New arene–ruthenium(ii) complexes with pyrazole derivatives as ligands were synthesized and characterized. This compounds exhibited good cytotoxic effects, and strong biostatic and biocidal activity.

“…Ligands a and b from which the complexes were derived also exhibited modest antiproliferative activities towards all tested cancer cell lines (Table 1), however, to a lesser extent than their chlorido complexes. The ruthenium p-cymene precursor [Ru(p-cymene)Cl 2 ] 2 of complexes 1a and 1b is known to be inactive towards cancer cells [22,[55][56][57][58], and our group determined its activity on A2780 and A549 cells with similar results (Figure S15). Hence, the enhancement of ligand anticancer activity can be attributed to the formation of the new complex.…”

“…Whereas, the Ru(III/IV) complex activity remains at the same level regardless of the concentration used (74%). The Ru(II) complex is characterized by anti-biofilm activity amount of 46% (at concentration of 0.5 mM) despite the fact that arene-ruthenium complexes containing p -cymene ring are described in the literature as compounds with a very high antimicrobial and anticancer activities [ 45 , 46 , 47 , 48 ]. The lowest percent inhibition of biofilm formation is observed in the presence of the Ru(III) complex.…”

“…[17][18][19] Theoretical studies using DFT The results of density functional theory (DFT) calculations verified the structures obtainedf rom single-crystal XRD analysisa s well as the diamagnetic character of Ru II in the studied complexes (see Figures S69-S71 andT ables S5 and S6 in the Supporting Information). [25][26][27][28][29][30] The electrostatic potential( ESP) surface diagrams (see Figure S70) revealed that the electron density is highest on the two chloride ligands, the carbonyl oxygen and the oxygen of furan. [27,28] The effect of the electron-donating/withdrawing substituents on the furoylthiourea ligand is mostly observed as al ocalized effect.…”
