Antibody to Collagen Type I in Periodontal Disease

Abstract: Serum antibody levels to human collagen Type I were measured in 97 patients with periodontal disease (Russell Periodontal Index 1.0-7.0) and 57 control subjects (Periodontal Index less than 1.0) using an enzyme-linked immunosorbent assay, which had been standardized with antisera prepared against human collagen Type I in rabbits. Absorption studies were used to confirm the specificity of antibodies to human Type I collagen. Levels of antibody to Type I collagen detected in patients were higher (P less than 0.0… Show more

“…Anti-collagen antibody has been consistently detected and suggested to be involved in gingival tissue destruction. 11,12,37 In addition to their well-known role in antibody production, B cells also have the determinative role as antigen-presenting cells (APCs) in the generation of T-cell-mediated immune responses. 38 Recent studies have shown that B cells are critical in the pathogenesis of T-cell-mediated autoimmune diseases.…”

“…In fact, much of this non-protective antibody may be locally produced auto-antibodies against gingival tissue components (collagen). 11,12 B-cell regulatory cytokines also play an important role in B-cell activation, and many have been found present in high levels in inflamed gingival tissues and in gingival crevicular fluid from periodontitis patients. Studies by Fujihashi et al 13 demonstrated high levels of IL-5 and IL-6 mRNA expression in gingival mononuclear cells isolated from severe periodontitis patients, but IL-2 and IL-4 mRNA expression was not detected.…”

“…Although most of the research has focused on anticollagen antibodies, many of the reports are contradictory. For instance, although Ftis et al 14 reported that the level of antibodies to collagen type I in peripheral blood was significantly higher in patients with periodontitis than in healthy controls, Hirsch et al 10 reported that anticollagen‐producing cells were rarely detected in peripheral blood and levels of anticollagen antibodies in serum were low in patients with periodontitis. At the same time, Sugawara et al 15 compared anticollagen IgG levels in the GCF and serum of patients with periodontitis; they reported that the IgG levels in GCF were slightly higher than those in autologous sera compared to healthy control subjects.…”
