Antibody responses to mumps virus proteins in natural mumps infection and after vaccination with live and inactivated mumps virus vaccines

Abstract: Paired sera from 20 patients with acute mumps infection, 16 from persons vaccinated with live attenuated mumps virus vaccine, and 12 from persons vaccinated with formalin-inactivated virus vaccine were studied for mumps antibodies by single radial hemolysis (SRH), hemagglutination inhibition (HI), and by enzyme immunoassays (EIA) specific for whole virus, envelope glycoprotein, and nucleocapsid antibodies. Mumps patients had diagnostic rises in serum mumps antibodies in 90-100% of the cases depending on the me… Show more

“…High IgM responses were slightly more common in the seronegative gruop, which was also observed by IIonen et al, (1 1). This is in accordance with the finding that previously vaccinated patients develop low IgM responses during mumps infection (12,20).…”

“…We demonstrated that current ELISAs could not reliably diagnose mumps at this stage of illness in this cohort of patients. Previous studies on mumps-IgM ELISAs (Gut et al, 1985;Julkunen et al, 1984;Meurmann et al, 1982;Popow-Kraupp, 1981; reported significantly higher sensitivities, achieving positive results with 75-98% of the acute sera tested. The assays used in the 1980s were generally indirect in-house ELISAs using crude antigen preparations.…”

“…Virus culture and IgM-ELISAs are the mainstay of diagnosis for many laboratories. In the 1980s, the mumps-IgM ELISA was shown to be superior to indirect the IgM-immunofluorescence test (IF), haemagglutination inhibition (HI), and complement fixation test (CFT) in diagnosing recent infection (Gut et al, 1985;Julkunen et al, 1984;Meurmann et al, 1982;Popow-Kraupp, 1981;. Information on the performance of mumps-IgM ELISAs in the current epidemic in the UK is 1386-6532/$ -see front matter © 2006 Elsevier B.V. All rights reserved.…”

“…Serum samples. Paired sera from 12 patients with acute parotitis or meningitis were serologically tested for mumps infection by single radial haemolysis [10]. The first sample from each patient was obtained 1-7 days after the onset of symptoms, the second sample 11-30 days later.…”

“…Serum samples were obtained on the day of the first vaccination and 5 weeks later. Fifteen subjects vaccinated with live vaccine and seven subjects vaccinated with inactivated virus vaccine were found to be seronegative for mumps antibodies before vaccination [10] and the pairs from these individuals were included in the study.…”
