Antibody‐induced activation of the hyaluronan receptor function of CD44 requires multivalent binding by antibody

Abstract: CD44 can function as a receptor for hyaluronan (HA). However, many cell lines and normal hematopoietic cells that express CD44 do not constitutively bind HA. A monoclonal antibody (mAb) specific for CD44 (IRAWB 14) has been described previously which induces CD44-mediated binding of HA rapidly (seconds to minutes) in some cell lines and in normal murine T cells. Of 16 CD44-specific mAb tested in the present study, only 3 exhibited this activity. Monovalent Fab fragments were prepared from two IgG2a antibodies … Show more

“…The mArg41 mutant did not bind HA at all, a finding that is also consistent with previous reports (40,41). Although binding of FL-HA to the tailless transfectant increased dramatically in the presence of "inducing" mAb IRAWB14 (29,42), no ligand binding to the mArg41 mutant could be induced by IRAWB14 treatment (results not shown).…”

“…CD44 clustering or dimerization seems required for HA binding in some cell types, particularly in lymphocytes, as demonstrated by, e.g. the effect of multivalent but not monovalent CD44 antibodies (56,57). CD44 clustering may also be required for ERM activation (41).…”

“…Hyaluronan from rooster comb (Sigma) was fluorescein-conjugated with fluoresceinamine (46). Cellsurface binding of fluorescein-conjugated HA (FL-HA) was determined by flow cytometry (47). HA oligosaccharides HA 4 , HA 6 , HA 8 , HA 10 , HA 12 , HA 14 , and HA 16 were made as described (48).…”
