Antibodies to a continuous region at residues 38-54 of hen egg white lysozyme found in a small fraction of anti-hen egg white lysozyme antibodies

Abstract: The antigenic reactivity of the region at residues 38-54 of hen egg white lysozyme (lysozyme) was investigated by using various rabbit, goat, and sheep anti-lysozyme antisera by the charcoal-dextran method. This continuous antigenic region was completely independent in its specificity from other discontinuous antigenic regions of lysozyme. The antibodies to this region were shown to be reactive with native lysozyme as well as thermolytic nondisulfide peptides (residues 38-54 and 38-57) and synthetic peptides (… Show more

“…All of these mouse strains are high responders to HEL and indeed anti-HEL antibodies could easily be isolated by HEL coupled to Sepharose CL-4B. However, in sharp contrast to our earlier studies in rabbits (8,9,23,32), sheep (22,32), and guinea pigs (10, 24), we have so far not succeeded in isolating HEL-region-specific antibodies by utilizing peptide (M.W. 4,000-5,000) affinity chromatography, presumably because of the low affinities of the peptide-antibody reactions.…”

“…The three discontinuous lysozyme sites account for more than 98% of anti-lysozyme antibodies raised in several species (Atassi, 1978;Atassi, 1979). A possible minor determinant, accounting for less than 1% of the total antibody, has also been identified (Takagaki et al, 1980). The participation of the surface-simulation sites in T cell recognition, however, has not previously been investigated.…”

“…Also trace amounts of antibodies have been reported [92] to a continuous region of hen egg-white lysozyme (residues 25-54) that account for about 0.10-1% of the total antilysozyme antibodies. In very recent studies [lo -121, employing the overlapping synthetic peptide approach, peptides that were seemingly inactive in binding of '251-labelled anti-Hb antibodies in a titration assay were further studied by attempting to isolate, from adsorbents of these peptides, antibodies specific for Hb.…”
