Antibodies as a Source of Anti-Infective Peptides: An Update

Abstract: This review focuses on antibodies (Abs) and their function in immune protection, with particular emphasis on microbicidal Abs. Some aspects of Abs and Ab-drug conjugates as targeting therapeutic agents are also discussed. The main aim, however, is devoted to Ab-derived peptides modulating functions of the immune system and to the latest experimental evidence of Abs as a source of anti-infective and antitumor peptides derived from their complementarity determining regions and constant regions.

“…Overall, peptides encoded by Ig genes exerted an effective antifungal activity. Other possible biological functions (antibacterial, antiviral, anti-tumour, immunoregulatory), already recognised for previously described Ab-derived peptides 4 , 5 , 7 , 8 , could be investigated in future studies.…”

“…In our previous works, we demonstrated that synthetic peptides related to CDRs and domains of the constant regions of Abs of different classes may display antimicrobial, antiviral, immunomodulatory and/or anti-tumour activities, mediated by different mechanisms of action, entirely unrelated to the specificity of the originating Ab 4 8 .…”

“…Antimicrobial cryptides have been identified in common mammalian proteins, as hemoglobin, albumin, immunoglobulins, lactoferrin and salivary proteins, as well as in plant ribosome inactivating protein [21][22][23]. Our research group focused, in particular, on the anti-infective properties of synthetic peptides whose sequence represents fragments of variable and constant regions of antibodies (Ab) [24]. Several years ago, the work started from the decapeptide AKVTMTCSAS (A10S, killer peptide KP).…”
