Antibodies as a Probe in Cytochrome P450 Research

Abstract: ABSTRACT:Cytochrome P450 (P450) is the superfamily of enzymes responsible for biotransformation of endobiotics and xenobiotics. However, their large isoform multiplicity, inducibility, diverse structure, widespread distribution, polymorphic expression, and broad overlapping substrate specificity make it difficult to measure the precise role of each individual P450 to the metabolism of drugs (or carcinogens) and hamper the understanding of the relationship between the genetic/environmental factors that regulate… Show more

“…Given the marked C-terminal amino acid variation and its hydrophilicity the use of peptides to the C-terminus of individual P450 as immunogens to produce monoclonal antibodies to individual forms of P450 has proven for many research groups including our own to be a highly efficient strategy to develop individual form-specific P450 monoclonal and polyclonal antibodies [30], [61], [62].…”

“…However, there were no commercial antibodies available, thus we prepared the rabbit polyclonal antibodies against it. The anti-peptide approach to antibody production overcomes the need to use purified P450s as antigens and is considered as a relatively simple, rapid, and effective method of producing antibodies [ 39 ]. Moreover, a high degree of binding specificity can be achieved by directing anti-peptide antibody toward unique regions of CYP6AA9.…”

“…Based on our experience, the first two types of studies are the most straightforward, especially when multiple CYP isoforms are involved. Antibody inhibitors and/or chemical inhibitors can be used in incubations with HLM, however, even though a number of CYP inhibitory antibodies are commercially available (Shou and Lu 2009;Tang et al 2005), they are not as commonly used as chemical inhibitors.…”
