Resistance of microorganisms to available antimicrobial agents have resulted in pernicious effect to human health over the last decades. The increase in antimicrobial resistance of pathogenic microorganisms particularly in tap surfaces is a major concern to the society. This research focused on the investigation of antimicrobial resistance pattern of pathogenic organisms (bacteria and fungi) isolated from various tap surfaces in the Federal University of Technology Akure (FUTA) Obakere campus, in Akure, Ondo State, Nigeria. Samples were obtained from different locations (laboratories, hostels and toilets) in FUTA. The samples were analyzed microbiologically on selective, differential and general purpose media. The isolated organisms were identified by the routine microbiological methods. Among the pathogenic microorganisms isolated were Pseudomonas aeruginosa, Klebsiella pneumoniae, K. oxytoca Escherichia coli, Bacillus cereus, Citrobacter freundii, Acinectobacter baumannii, Staphylococcus aureus, Aspergillus fumigatus, Aspergillus niger, Candida albicans and Fusarium oxysporium. The percentage distribution varies with P. aeruginosa (26.09%) having the highest percentage and Klebsiella sp. (4.45%) recorded at the lowest percentage of occurrence. All fungi isolated have simillar percentage distribution. The isolates displayed various levels of resistance to piperacillin (100%), ampicillin (66.67%). ceftazidime (66.67%), tetracycline (83.33%), cefepime (66.67%) and chloramphenicol (66.67%). The resistance to amoxicillin-clavulanate by E. coli and C. freundii isolated from taps surfaces were 60% and 50%, respectively. K. pneumoniae from tap water surface has 100% resistance to cefepime. The high resistance of microorganisms to antimicrobial agents indicated a great threat to people living within FUTA community that constantly obtaining water from these taps surfaces.

BackgroundPersistence of antibiotic resistant bacteria, including multidrug resistant (MDR) pseudomonads, is an important environmental health problem associated with drinking water distribution systems (DWDS) worldwide. There is paucity of data on the molecular characteristics of antibiotic resistance genes and their mode of transfer among pseudomonads from DWDS located in resource-challenged areas such as southwestern Nigeria.MethodsMDR pseudomonads (n = 22) were selected from a panel of 296 different strains that were isolated from treated and untreated water in six DWDS located across southwest Nigeria. Primarily, the isolated pseudomonads strains were identified by 16S rDNA sequencing and antibiotic-resistance testing was completed using agar breakpoints assays. The final panel of strains of resistant to more than three classes of antibiotics (i.e. MDR), were further characterized by PCR genotyping, Sanger sequencing, and plasmid profiling.ResultsPseudomonad resistance to gentamicin and streptomycin ranged from 22.7 to 54.6 % while resistance to tetracycline, ceftiofur and sulphamethoxazole ranged from 40.9 to 77.3 %. The most commonly detected antibiotic resistance genes were tet(A) (31.8 % of isolates), sul1 (31.8 %), blaTEM (40.9 %) and aph(3″)c (36.4 %). Class 1 integron sequences were evident in 27.3 % of isolates and they harbored genes encoding resistance to aminoglycosides (aadA2, aadA1), trimethoprim (dfrA15, dfr7) and sulphonamide (sul1) while the plasmid ranged between 22 and 130 kb.ConclusionsPseudomonas spp, isolated from these DWDS possess resistance genes and factors that are of public and environmental health significance. Therefore, has the potential of contributing to the global scourge of resistance genes transfer in human, animals and environments, thereby, useful in the epidemiology of antimicrobial resistance.

Aims: The current investigation stands for the determination of MAR (multiple antibiotic resistance) indices of antibiotic resistant and heavy metal (HM) tolerant bacterial isolates from Mahananda river water near Malda (West Bengal state, India) and profiling of R-plasmids of the isolated bacteria. Methods: The water samples (n=5), collected from Mahananda river water, Malda town, (India), were checked for the presence of bacterial growth in nutrient broth cultures. The pure bacteria colonies, procured from each of the nutrient broth cultures, were identified by conventional methods. The susceptibilities to antibiotics and HMs of the isolated bacteria were determined by disc diffusion and agar dilution, respectively. Plasmid curing of the resistant bacteria was done with SDS treatment, and the isolated plasmids were screened through agarose gel electrophoresis. The MAR indices for the isolates were calculated. Results: The isolated bacteria were identified as Escherichia coli (n=3), and Pseudomonas aeruginosa (n=2). The bacterial isolates showed various patterns of resistance to antibiotics and heavy metals. The MAR indices for the bacterial isolates ranged 0.0-0.2, for E. coli, and the value was 0.47, for both the Ps. aeruginosa isolates. The isolated bacteria harboured a single plasmid mediating co-resistance to antibiotics and HMs. Conclusion: The current study demonstrates the occurrence of plasmid, encoding Am-Cm-Ce-Cx-Tm-Cd 2+-Hg 2+ and Cm-Tm-Cd 2+-Hg 2+ resistances, among the aquatic bacteria of Mahananda river that may potentially act as the source of dissemination of pathogenic bacteria and bacterial antibiotic resistances, requiring public awareness on the issues of misuse and/or overuse of antibiotics.
