Antibiotic polymyxin arranges lipopolysaccharide into crystalline structures to solidify the bacterial membrane

Abstract: Polymyxins are last-resort antibiotics with potent activity against multi-drug resistant pathogens. They interact with lipopolysaccharide (LPS) in bacterial membranes, but mechanistic details at the molecular level remain unclear. Here, we characterize the interaction of polymyxins with native, LPS-containing outer membrane patches of Escherichia coli by high-resolution atomic force microscopy imaging, along with structural and biochemical assays. We find that polymyxins arrange LPS into hexagonal assemblies t… Show more

“…More care would need to be taken for labeling Gram-negative bacteria to ensure that the used fluorescent marker labels only the membrane of interest, either the inner or the outer membrane. Third, additional care must be taken when performing FCS experiments in Gram-negative bacteria, as the topography of the outer membrane was previously shown to be affected by various compounds (26, 64) which itself affects measurement of diffusion speeds with FCS (65). Our work confirmed the results of previous experiments on the effect of a cold shock on membrane fluidity.…”

“…Nevertheless, improved dosing regimens and the rise of Gram-negative MDR strains led to a renaissance of their clinical use when everything else was failing 4 . Sadly, PM resistance has also emerged, mostly due to outer membrane lower permeability 7 , but also through nonspecific mechanisms (e.g.…”

“…In many organisms, like Escherichia coli, the lipid A portion of LPS is negatively charged due to the presence of phosphate groups on the glucosamine disaccharide (3). These phosphates are the binding sites for cationic antimicrobial peptides (CAMPs) like polymyxins (4,5). While LPS is generally considered to be essential, LPS-null mutants of several Gram-negative organisms have been isolated including Acinetobacter baumannii (6), Moraxella catarrhalis (7), Neisseria meningitidis (8), and Caulobacter crescentus (9).…”
