Antibacterial activity of microbicidal cationic proteins 1 and 2, natural peptide antibiotics of rabbit lung macrophages

Abstract: Microbicidal cationic proteins 1 and 2, peptides derived from rabbit lung macrophages, were tested for bactericidal activity against various bacterial species. Both were highly active against diverse gram-positive and gram-negative organisms under conditions of near-neutral pH (between 7 and 8) and relatively low ionic strength. Susceptible species included Staphylococcus aureus, Staphylococcus epidermidis, Streptococcus pneumoniae, Listeria monocytogenes, Pseudomonas aeruginosa, Klebsiella pneumoniae, Haemoph… Show more

“…Interestingly, human alveolar macrophages expressed mRNA for hBD-3 upon activation with IFN-c and LPS. Our observation that alveolar macrophages, but not monocytes and MDM, express a particular antimicrobial peptide is certainly not unique, since alveolar macrophages in rabbits 45 produced antimicrobial peptides not found in other types of mononuclear phagocytes.…”

“…Bactericidal kinetics was determined using a modified version of the assay described by Lehrer et al [20]. CP1 at the MIC concentration of 3 µM was incubated with 10 5 CFU/ml for 30 min.…”

“…First characterized as granule components of phagocytic leukocytes (25), defensins are 3-4 kDa cationic peptides with well-characterizedantimicrobialactivities (26). Six invariantly placed cysteine residues form disulfide bonds between Cys'-Cys6, Cys2-Cys4, and Cys3-Cys5 (see Fig.…”
