“…The mAbs were specific for families of TCR-chain variable regions, indicating clonality of a T-cell population in lymphomas (Clark et al, 1986). After using mAbs on CTCL biopsies, clonal populations in the peripheral blood of SS patients was indicated (Charley et al, 1990) and confirmed by several groups (O'Grady et al, 1990;Heald et al, 1994;Kuchnio et al, 1994;Bogen et al, 1996;Dummer et al, 1996) In order to monitor treatment responses, we studied the level of clonal T cells in CTCL patients and the clinical follow-up during different therapies. We found an association between CD4 + Vb + clonal levels and the clinical development during treatment.…”

“…In an inflammatory infiltrate, no more than a few percent of T cells will react with any one anti-V~ gene family monoclonal antibody. In contrast, in a T cell lymphoma or leukemia all of the tumor cells will react with the particular anti-V~ antibody that is specific for the V~ gene that they express [6,7].…”

“…Two forms of TCR exist, namely TCR α/β and TCR γ/δ. Several monoclonal antibodies recognizing framework determinants of TCR α/β and ΤCR γ/δ by immunohistochemistry and flow cytometry have been described [2][3][4]. In particular the T cell receptor α/β is expressed inconsistently by malignant cell populations and these aberrant immunophenotypic expressions are highly suggestive of a T cell clonal population [2].…”

“…Monoclonal antibodies have been produced which recognize families of many of these variable regions (particularly the beta chain). The finding of an excess of 1 family of TCR-beta chain rearrangement not only may assist in the diagnosis of a clonal lesion, but may also be used to monitor the efficacy of therapy against the clone [4]. A limitation on the use of these reagents is the relatively large panel of monoclonal antibodies that is necessary to encompass the entire TCR repertoire.…”
