Background and objective: The purpose of the current investigation was to study the influences, preventive and diuretic, of Nigella sativa L. seeds oil (NSSO) on calcium oxalate (CaOx) urolithiasis induced in Wistar male rats. Methodology: Seeds of Nigella sativa L. (N.S) were analysed for the evaluation of the concentration of oxalate and calcium. Nigella sativa L. seeds oil is obtained by hydrodistillation and HPTLC densitometric method was adopted to determine the amount of thymoquinone (TQ) present. Thirty male Wistar rats were divided into 5 groups (N=6). Group I, negative control, drank tap water. The other groups were II Positive control, III, IV and V received a treatment model inducing calcium oxalate urolithiasis for 28 days, using an aqueous solution involve 0.75% (EG) ethylene glycol and 1.0 % (AC) chloride ammonium. Rats in group III received in addition, 750 mg/kg Cystone from the beginning to the end of calculi induction experimentation. However, rats in Groups IV and V received (NSSO) at 5 ml/kg b.w by gavage on days 1 st to 28 th and 15 th to 28 th days, respectively. On days 0, 7, 14, 21 and 28, body weights were measured and the 24-hour urine samples were accumulated and analysed for biochemical elements. On the 28 th day, blood samples were collected for the estimation of serum parameters including creatinine, BUN and uric acid. All animals were sacrificed at the end of the experiment and the kidneys were detached for histopathological examination. Results: Administration of (NSSO) at 5 ml/kg body weight/dose/day for 28 days exerts a protective effect by reducing significantly (p <0.01) urinary and serum rates of calcium, phosphate and oxalate. This preventive diet could increase the volume of urine excreted. Conclusion:The nephroprotectrice and diuretic activity demonstrated by Nigella sativa L. gives a scientific basis that approves their traditional use like a remedy against urolithiasis.

Context: Different parts of Punica granatum Linn. (Punicaceae) are traditionally used as renal protective agents in the Indian system of medicine. However, there is paucity of information regarding its role in diabetic nephropathy. Objective: The present study investigates the nephroprotective potential of flavonoid-rich fraction of P. granatum leaves in streptozotocin (STZ)-induced early diabetic nephropathy in experimental animals. Materials and methods: Experimental diabetic nephropathy was induced in Wistar rats by single intraperitonial injection of STZ (65 mg/kg) dissolved in ice cold citrophosphate buffer (pH 4.3). After induction rats were divided into five groups (6 normal; 24 diabetic) and administered with glibenclamide (5 mg/kg) and three dose levels of flavonoid-rich fraction of P. granatum leaves (PGFF), i.e. 50, 100, and 200 mg/kg body weight/day for 28 d. Fasting blood glucose, lipid profile, serum albumin, serum total protein, serum creatinine, blood urea nitrogen (BUN) glycosylated hemoglobin, and biomarkers of kidney oxidative stress were assessed at the end of the treatment period. Urine was analyzed for the measurement of total protein, albumin, and creatinine clearance. Kidney sections were subjected to histopathological study. Results: Daily oral administration of variable dose levels of PGFF for 28 d normalized various biochemical, metabolic, and histopathological changes in the diabetic rats. PGFF significantly (p50.01 and p50.05) improved the glycemic status and renal function in diabetic rats as compared with diabetic control rats. Discussion and conclusion: The results of our study thus prove the protective effect of PGFF in early diabetic nephropathy by ameliorating proteinuria and disturbed glucose homeostasis in experimental animals.

Objective: The present study was aimed to evaluate the antiurolithiatic potential of the ethanolic extract of the seed of Phaseolus vulgaris (EPV). Materials and Methods: Calcium oxalate urolithiasis in male Wistar rats was induced by ethylene glycol (EG) (0.75% v/v) and ammonium chloride (1% w/v) administration in drinking water. Cystone (750 mg/kg, p.o.) was used as a standard drug, and EPV was administered at doses of 200 and 400 mg/kg, p.o. Both preventive and curative effects of EPV were evaluated. Urinary biochemical parameters such as calcium, oxalate, phosphate, uric acid and creatinine; deposition of calcium and oxalate in the kidney; and serum uric acid, creatinine, and blood urea nitrogen (BUN) were assessed. Creatinine clearance was calculated. Oxalate associated oxidative stress in the kidney was assessed by estimating in vivo antioxidant parameters such as lipid peroxidation, superoxide dismutase, catalase, and reduced glutathione. Histopathological studies of the kidney were carried out. Results: In the preventive and curative disease-control groups, urinary excretion of calcium, oxalate, and their deposition in the kidney were significantly increased. Elevated levels of phosphate and uric acid in urine and uric acid, creatinine, and BUN in serum were observed in both the control groups. Creatinine clearance was reduced in the control groups. On treatment with cystone and EPV, all the urinary, serum biochemical, and oxidative stress parameters were reversed to almost normal values. Cystone and EPV significantly restored the in vivo antioxidant enzymes by decreasing the lipid peroxidation in the kidney. This study demonstrated the antiurolithiatic activity of the ethanolic extract of P. vulgaris seeds against EG-induced renal calculi in Wistar rats.
