Objective: The present study was carried out to investigate the role of curcumin in osteoarthritis when it is used as an adjuvant to diclofenac sodium.Methods: Osteoarthritis (OA) was induced by administering nalidixic acid 400 mg/kg. Animals (Female rats) were divided into 5 groups each containing 6 animals. Group I was considered as control, group II in which the animals were induced with osteoarthritis with nalidixic acid and were given no treatment. Group III in which osteoarthritis induced animals were treated with diclofenac sodium by the oral route. Group IV osteoarthritis induced animals were treated with the combination of diclofenac sodium and curcumin and group V animals were pre-treated with curcumin and then induced with osteoarthritis. Parameters like ulcer area, ulcer index, free acidity, total acidity, the volume of gastric juice were estimated. Histopathological studies were also carried out.Results: The data of our study shows that nalidixic acid has not shown much effect on the gastric parameters in group II animals. The ulcer index, free acidity, total acidity and gastric juice volume were increased significantly (p<0.001) in group III and decreased in group IV animals (p<0.05) when compared to group I control animals. Group V animals pretreated with curcumin have shown fewer incidences of gastric ulcers and other ulcerative parameters non significantly. Histopathology also suggests a low incidence of ulcers in group IV and group V.Conclusion: It demonstrates that curcumin when used along with the conventional NSAIDs as an adjuvant therapy, has a role in treating osteoarthritis effectively.

Peptic ulcers are an important pathology, and the search for safer and more effective treatment methods is of paramount importance. In this study, we assess the gastroprotective effects of the hydroethanolic extract (HE) and ethyl acetate fraction (EAF) from Kalanchoe pinnata leaves against an ethanol/HCl-induced ulcer model in rats. The HE reduced gastric lesions by approximately 47% (400 mg/kg). A significant inhibition of the gastric lesions by 50% was observed after pretreatment with the EAF (200 mg/kg). Quercetrin and quercetin 3-O-α-L-arabinopyranosyl-(1→2)-α-L-rhamnopyranoside were isolated and identified in the flavonoid fraction (EAF) by HPLC and NMR analyses because this fraction showed the highest gastroprotective effect. This fraction demonstrated high antioxidant activities (CE 50 =41.91 µg/mL) by DPPH in comparison with Trolox ® and 11.33 mmol Trolox ® equivalent by ORAC. In conclusion, the HE and FAE from K. pinnata displayed gastroprotective activity in rats, most likely due to the presence of flavonoids.Uniterms: Gastric ulcer/study/rats. Gastric ulcer/treatment. INTRODUCTIONFor the past two centuries, peptic ulcers have caused high morbidity and mortality (Malfertheiner, Chan, Mccoll, 2009). This disease is a serious and prevalent pathology (Calam, Baron, 2001). A peptic ulcer is a chronic lesion, usually single, that can be located in the gastric or duodenal mucosae (Kumar, Abbas, Fausto, 2005). In the UK, the incidence of this disease was 12% higher in men than in women (Araujo, Borini, Guimarães, 2014). A review article (January 1980 to February 2009 showed that the incidence rate of uncomplicated peptic ulcers was approximately one case per 1000 people per year in the general population; the incidence rate for complicated peptic ulcers that included bleeding and perforation was approximately 0.7 cases per 1000 people per year (Lin, Rodrígues, Hernándes-Diaz, 2011).The pathophysiology of peptic ulcers suggests that this disease is caused by an imbalance between defensive factors (mucus and bicarbonate secretion, mucosal barrier, blood flow, and prostaglandins) and destructive factors (reactive oxygen species and acid and pepsin secretion), resulting in the acute inflammation of the gastrointestinal mucosae (Laine, Takeuchi, Tarnawski, 2008). Several stimuli serve as offensive factors, including Helicobacter pylori, the use of nonsteroidal anti-inflammatory drugs (NSAIDs), stress, alcohol and cigarette smoking (Malfertheiner, Chan, Mccoll, 2009;Malnick et al., 2014). During the past years, evidence accumulated showing that glycine comprises anti-inflammatory activities. These effects occur, at least in part, via the activation of glycine-gated chloride channels (GlyR). Glycine is one of the major structural units of collagen, making up about 30% of the amino acids. This study aims to investigate the anti-inflammatory potential of collagen hydrolysate (CH) using the zymosan-induced ear-skin inflammation mouse model. After oral intake of 12.5, 25 or 50 mg CH the plasma levels of glycine increased in a concentration-dependent manner. CH was able to counteract zymosan-induced ear-skin inflammation locally (ear swelling) as well as systemically (IL-6 production by lipopolysaccharide (LPS)-stimulated whole blood cells). The LPS-stimulated IL-6 production in whole blood correlated positively with the ear swelling response. This correlation was abolished by strychnine (a glycine receptor antagonist), indicating the involvement of GlyR. Collectively, these data show that CH is able to modulate inflammatory responses both locally as well as systemically. This effect might be constituted by inhibiting pro-inflammatory cytokine production via GlyR.
