Anti-tumor Synergistic Effect of a Dual Cancer-Specific Recombinant Adenovirus and Paclitaxel on Breast Cancer

Abstract: This study aimed at investigating the anticancer potential of the recombinant adenovirus Ad-apoptin-hTERTp-E1a (Ad-VT) and its synergistic combination with paclitaxel (PTX) in breast cancer treatment. First, we used the Calcusyn software to analyze the synergy between the Ad-VT and paclitaxel, and to determine the final drug concentration. Second, we used crystal violet staining and WST-1 assays to analyze the inhibitory effect of Ad-VT and paclitaxel combination treatment on MCF-7, MDA-MB-231, and MCF-10A cel… Show more

“…With advances in molecular biology and gene engineering technologies, many novel treatment schemes have been developed, including gene targeting and OV therapy. Compared with other cancer treatment methods, OVs have a broad antitumor spectrum, are safe and reliable, are less toxic with fewer adverse effects, and exert synergistic effects with other anticancer drugs [24]. Therefore, the development and clinical application of adenoviruses as drug vectors have been widely studied, and progress has been made.…”

“…The cell invasion was observed with a transwell assay according to a previous report. 40 Briefly, 200 μL of SMMC7721 or LO2 hepatocyte suspension was placed in the upper chambers coated with matrigel (1:7 with cell medium). Into the lower chambers, 800 μL of medium containing NETs or diabetic serum was added as a chemo‐attractant.…”
