Oxadiazole, a heterocyclic nucleus has attracted a wide attention of the chemist in search for the new therapeutic molecules. Out of its four possible isomers, 1, 3, 4-oxadiazole is biologically active, synthetically useful and widely exploited for various applications. Various biological activities are reported to be associated with 1, 3, 4-oxadiazoles. Encouraged by these reports the present study has been undertaken. The newly synthesized compounds were confirmed from IR, 1 H NMR and mass spectral data. All the synthesized compounds were subjected for anti angiogenesis as well as anticancer activity. Compounds 7b and 10d showed very good anti angiogenesis and anticancer activity compared to all other analogues in the series.

In the current scenario, development of anticancer drugs with specific targets is of prime importance in modern chemical biology. Observing the importance of benzophenone and coumarin nucleus, it would be worthwhile to design and synthesize novel benzophenone derivatives (8a-o) bearing the coumarin nucleus. Further, they were screened for prospective anticancer activities in vitro against the Michigan Cancer Foundation-7 (MCF-7) and Ehrlich's ascites tumor (EAT) cell lines and their biomarkers, followed by in silico studies regarding phosphoinositide 3-kinase (PI3K) and caspase by molecular docking. Benzophenones have been reported as potential drugs targeting tumor angiogenesis; thus, the formation of neovessels in an in vivo model system like CAM, which is angiogenesis dependent, was observed in the presence of compounds 8a-o. The above findings would help in understanding their putative potential as therapeutic agents for cancer patients.

Synthesis of Benzophenones and in vitro Evaluation of Their Anticancer Potential in Breast and Prostate Cancer Cells

Breast and prostate cancers are frequently treated with chemotherapy. Several novel chemicals are being reported for this purpose, particularly synthetic and natural benzophenones. This work reports the synthesis of substituted 2‐hydroxybenzophenones through 1,4‐conjugate addition/intramolecular cycloaddition/dehydration of nitromethane on key intermediate chromones. Structures were extensively studied by means of 2D NMR spectroscopy and single‐crystal XRD. Their cytotoxicity was evaluated in vitro in two breast cancer cell lines (MDA‐MB‐231 and T47‐D) and one prostate cancer cell line (PC3). The most potent compound exhibited good cytotoxic effects against the three cancer cell lines (IC50 values ranging from 12.09 to 26.49 μm) and induced cell‐cycle retardation only on prostate cancer cells, which suggested that it might exert cell‐type‐specific effects.
