Anti-Thymocyte Globulin Treatment Augments 1,25-Dihydroxyvitamin D3 Serum Levels in Patients Undergoing Hematopoietic Stem Cell Transplantation

Abstract: Application of anti-thymocyte globulin (ATG) is a widely used strategy for the prevention of graft-versus-host disease (GvHD). “…The study by Macedo et al [33] accurately described the immunomodulatory role of vitamin D, reporting that high levels may downregulate T cell proliferation via epigenetic gene modulation (EZH2 and ID3), whereas vitamin D deficiency may induce high production of proinflammatory cytokines such as tumor necrosis factor α (TNF-α), interleukin 2 (IL-2), IL-4, or interferon γ (INF-γ). Furthermore, vitamin D could have a synergistic action with antithymocyte globulin (ATG) serum administered to prevent GvHD: in vitro , ATG increases gene expression of CYP27B1, the enzyme responsible for the conversion of 25-hydroxyvitamin D3 into 1,25-dihydroxy vitamin D3, known to induce tolerogenic dendritic cells and T regulatory cells [34].
