Anti-tax interacting protein-1 (TIP-1) monoclonal antibody targets human cancers

Abstract: Radiation-inducible neo-antigens are proteins expressed on cancer cell surface after exposure to ionizing radiation (IR). These neo-antigens provide opportunities to specifically target cancers while sparing normal tissues. Tax interacting protein-1 (TIP-1) is induced by irradiation and is translocated to the surface of cancer cells. We have developed a monoclonal antibody, 2C6F3, against TIP-1.Epitope mapping revealed that 2C6F3 binds to the QPVTAVVQRV epitope of the TIP-1 protein. 2C6F3 binds to the surface … Show more

“…Flow cytometry was performed as described earlier [36]. Cells were either sham irradiated or irradiated with 3Gy and harvested at 24, 48, 72 and 96 h. Monoclonal antibodies against human TAF15 and isotype control were purchased from Abcam, USA.…”

“…These radiation inducible genes may also serve as antigens for antibody targeted therapy. Yan et al has developed a monoclonal antibody, 2C6F3, that recognizes radiation inducible Tax interacting protein-1 (TIP-1) (30). Binding of 2C6F3 to irradiated cancer cells promoted antibody dependent cell-mediated cytotoxicity and phagocytosis and a radiolabeled 2C6F3 inhibited tumor growth in vivo .…”

“…TIP1 was initially identified as one of the binding partners of the T-cell leukemia viral oncoprotein Tax [9]. A single PSD-95/DlgA/ZO-1 (PDZ) domain (89 amino acids) is the only structural and functional unit in the small protein (total of 124 amino acids in human and mouse), distinguishing TIP-1 from other PDZ proteins [9,10]. The TIP1 expression is elevated in various cancers including lung, breast, and gliomas.…”

“…The TIP1 expression is elevated in various cancers including lung, breast, and gliomas. Radiation stress leads to induction of TIP1 within the cells, as well as the cell surface [7,10]. Cell-surface TIP1 is detected on many types of cancer cells in vivo and is not usually present on normal cells [7,10].…”
