Anti-Skin Inflammatory and Anti-Oxidative Effects of the Neoflavonoid Latifolin Isolated from Dalbergia odorifera in HaCaT and BJ-5ta Cells

Abstract: Skin is the first line of defense in the body against external stimulation and injury. Inflammation and oxidative stress in skin cells are the initiators and promoters of several skin diseases. Latifolin is a natural flavonoid isolated from Dalbergia odorifera T. Chen. This study aimed to evaluate the anti-inflammatory and antioxidant properties of latifolin. The anti-inflammatory effects were evaluated using tumor necrosis factor-α/interferon-γ (TNF-α/IFN-γ)-treated HaCaT cells, revealing that latifolin inhib… Show more

“…To further evaluate the in vitro biological activity of the GS and GF, we used two human cell lines, HaCaT (keratinocytes) and BJ (fibroblasts), which are commonly used to evaluate the effects of antioxidant compounds on skin [ 55 , 56 ]. Specifically, we conducted cell viability assays to determine the effect of the GS and GF on the growth of HaCaT and BJ cells.…”

“…Once activated, keratinocytes produce numerous cytokines, including IL-1β and IL-6, which intensify the inflammatory response through autocrine and paracrine actions [10,39]. Moreover, when HaCaT cells are co-stimulated with TNF-α and IFN-γ, it triggers the generation of Th-2-related chemokines such as CXCL8 and MDC [40,41]. These chemokines are crucial The HaCaT cells underwent a pre-treatment with 10 µM kahweol for 1 h, followed by stimulation using TNF-α/IFN-γ (10 ng/mL each) for 2 h. Nuclei were visualized using DAPI.…”

Moreover, when HaCaT cells are co-stimulated with TNF-α and IFN-γ, it triggers the generation of Th-2-related chemokines such as CXCL8 and MDC [40,41]. These chemokines are crucial in recruiting inflammatory cells to skin lesions, thus contributing to the progression from acute to chronic inflammatory responses [42].…”
