Anti-RO/SSA and anti-La/SSB antibodies: Association with mild lupus manifestations in 645 childhood-onset systemic lupus erythematosus

Abstract: Our large multicenter cohort study provided novel evidence in cSLE that anti-Ro/SSA and/or anti-La/SSB antibodies were associated with mild manifestations, particularly cutaneous and musculoskeletal. Secondary Sjögren syndrome was rarely observed in these patients, in spite of comparable frequencies of anti-Ro/SSA and/or anti-La/SSB reported for adult SLE.

“…Rates of anti-Ro antibodies present in cSLE patients range from 23-56% and anti-La antibodies range from 7-37% [21][22][23]. However, there are very few reports of secondary SS in cSLE, mostly con ned to case series [5][6][7]. Our imaging ndings may represent subclinical disease that could manifest itself with more overt clinical symptoms as time goes on.…”

“…To date, the relationship between anti-SSA(Ro)-antibodies and neurological manifestations in pSS is not sufficiently understood, in particular since both antibody-negative courses and isolated antibody-positive associated myelitis have been described [7,8,10,19,24,25]. Only a few study groups have investigated the impact of CSF-anti-SSA(Ro)-antibodies in patients with neurological involvement of primary rheumatological disorders such as pSS or systemic lupus erythematosus, reporting an intrathecal synthesis occurring partly prior to systemic manifestation [9,26].…”

“…In addition, SSB is one of the down-regulated hub genes in PPI module. Autoantibodies to the encoded protein are found associated with some other autoimmune disease including Sjogren syndrome27 and systemic lupus erythematosus 28. Therefore, the processes including cilium assembly and eNOS activation, and some genes including RABEP1 and SSB would participant in progressive fibrosis and autoimmune response of lcSSc.The pathways of NCAM1 interactions and CREB phosphorylation through the activation of CaMKII were enriched by up-regulated genes.…”
