Brunfelsia grandiflora is a South American solanaceae widely used since long ago for its recognized medicinal properties. We have recently reported its chemical composition, showing a relevant number of bioactive compounds with antioxidant capacity, and proved the cytoprotective and antioxidative stress potential of B. grandiflora in cultured endothelial cells. Since B. grandiflora extracts have shown effects on the central nervous system, the present study was designed to show the potential cytoprotective capacity and the antioxidative stress potential of phenolic extracts from the plant on cultured neuron-like cells, as a model to reduce the presentation or effects of chronic diseases of the nervous system. To this end, we studied its reactive oxygen species (ROS)-reducing capacity, its antioxidant defense mechanisms, and some molecular markers involved in redox balance and apoptosis. The results show that cell survival and most changes in biomarkers related to oxidative status, ROS, reduced glutathione, glutathione peroxidase and reductase, malondialdehyde, and caspase 3/7 activity, and molecular expression of cell death-related genes (BAX, BNIP3, and APAF1), NFκB, SOD, and NRF2 (genes from oxidative stress—antioxidants) induced by oxidative stress were prevented by either co- or pretreatment of neuron-like cells with B. grandiflora extracts (25–200 µg/mL). The results demonstrate the chemoprotective potential of the plant and support its medicinal use.

BackgroundThe preoperative inflammatory condition significantly influences the prognosis of malignancies. We aimed to investigate the potential significance of preoperative inflammatory biomarkers in forecasting the long-term results of lung carcinoma after microwave ablation (MWA).MethodThis study included patients who received MWA treatment for lung carcinoma from Jan. 2012 to Dec. 2020. We collected demographic, clinical, laboratory, and outcome information. To assess the predictive capacity of inflammatory biomarkers, we utilized the area under the receiver operating characteristic curve (AUC-ROC) and assessed the predictive potential of inflammatory biomarkers in forecasting outcomes through both univariate and multivariate Cox proportional hazard analyses.ResultsA total of 354 individuals underwent MWA treatment, of which 265 cases were included in this study, whose average age was 69.1 ± 9.7 years. The AUC values for the Systemic Inflammatory Response Index (SIRI) to overall survival (OS) and disease-free survival (DFS) were 0.796 and 0.716, respectively. The Cox proportional hazards model demonstrated a significant independent association between a high SIRI and a decreased overall survival (hazard ratio [HR]=2.583, P<0.001). Furthermore, a high SIRI independently correlated with a lower DFS (HR=2.391, P<0.001). We developed nomograms utilizing various independent factors to forecast the extended prognosis of patients. These nomograms exhibited AUC of 0.900, 0.849, and 0.862 for predicting 1-year, 3-year, and 5-year OS, respectively. Additionally, the AUC values for predicting 1-year, 3-year, and 5-year DFS were 0.851, 0.873, and 0.883, respectively.ConclusionSIRI has shown promise as a valuable long-term prognostic indicator for forecasting the outcomes of lung carcinoma patients following MWA.

Fish head byproducts derived from surimi processing contribute about 15% of the total body weight, which are beneficial to health because they contain essential nutrients. In this study, olive flounder (OF) was the target species in order to maximize the byproduct utilization. In RAW 264.7 macrophages, the seven hydrolysates from OF head byproducts were examined for their inhibitory potential against inflammation and the oxidative stress induced by lipopolysaccharides (LPS). The pepsin hydrolysate (OFH–PH) demonstrated strong anti-inflammatory activity via the down-regulation of NO production, with an IC50 value of 299.82 ± 4.18 µg/mL. We evaluated the inhibitory potential of pro-inflammatory cytokines and PGE2 to confirm these findings. Additionally, iNOS and COX-2 protein expressions were confirmed using western blotting. Furthermore, the results from the in vivo zebrafish model demonstrated that OFH–PH decreased the LPS-elevated heart rate, NO production, cell death, and intracellular ROS level, while increasing the survival percentage. Hence, the obtained results of this study serve as a platform for future research and provide insight into the mediation of inflammatory disorders. These results suggest that OFH–PH has the potential to be utilized as a nutraceutical and functional food ingredient.
