Despite the increasing interest in insect-based bioactive products, the biological activities of these products are rarely studied adequately. Larvae of Tenebrio molitor, the yellow mealworm, have been eaten as a traditional food and provide many health benefits. Therefore, we hypothesized that T. molitor larvae might influence adipogenesis and obesity-related disorders. In the present study, we investigated the anti-adipogenic and antiobesity effects of T. molitor larvae in vitro and in vivo. The lipid accumulation and triglyceride content in mature adipocytes was reduced significantly (up to 90%) upon exposure to an ethanol extract of T. molitor larvae, without a reduction in cell viability. Exposure also resulted in key adipogenic and lipogenic transcription factors. Additionally, in adipogenic differentiation medium the extract induced phosphorylation of adenosine monophosphate (AMP)-activated protein kinase and mitogen-activated protein kinases. Daily oral administration of T. molitor larvae powder to obese mice fed high-fat diet attenuated body weight gain. We also found that the powder efficiently reduced hepatic steatosis as well as aspartate and alanine transaminase enzyme levels in mice fed a high-fat diet. Our results suggest that T. molitor larvae extract has an antiobesity effect when administered as a food supplement and has potential as a therapeutic agent for obesity.

Seaweed, a popular and abundant food ingredient mainly consumed in Asian countries, is a good source of bioactive compounds with anti-obesity effects. However, the anti-obesity effects of Sargassum thunbergii have not yet been established. In this study, we isolated six indole derivatives (STCs)—indole-2-carboxaldehyde (STC-1), indole-3-carboxaldehyde (STC-2), indole-4-carboxaldehyde (STC-3), indole-5-carboxaldehyde (STC-4), indole-6-carboxaldehyde (STC-5), and indole-7-carboxaldehyde (STC-6)—from S. thunbergii and evaluated their inhibitory effects on adipocyte differentiation in 3T3-L1 cells. We found that STC-1 and STC-5 resulted in non-toxic inhibition of the differentiation of 3T3-L1 adipocytes and thus selected these compounds for further study. STC-1 and STC-5 significantly inhibited lipid accumulation and downregulated the expression of peroxisome proliferator-activated receptor-γ (PPARγ), CCAAT/enhancer-binding protein α (C/EBPα), and sterol regulatory element-binding protein 1c (SREBP-1c) in a dose-dependent manner. The specific mechanism mediating the effects of STC-1 and STC-5 was shown to be AMP-activated protein kinase (AMPK) activation. Our results demonstrated the inhibitory effect of STC-1 and STC-5 on adipogenesis through the activation of the AMPK signal pathway. Together, these findings suggested that STC-1 and STC-5 may be effective candidates for the prevention of obesity or obesity-related diseases.

BACKGROUND Microwave hydrodiffusion and gravity (MHG) of Sargassum muticum was proposed both for obtaining an aqueous extract and as pretreatment before ethanol extraction of bioactives. RESULTS No significant differences in composition and properties between the aqueous phase drained during MHG processing at 600 W/7 min (MHG‐1) and at 600 W/5 min + 300 W/5 min (MHG‐2) were observed. The aqueous extract, accounting for less than 0.2% of the initial material, was refined by adsorption onto food grade resins to recover and concentrate the phenolic fraction, further eluted in 68% ethanol. The final product contained more than 50% phlorotannins and the antiradical capacity was higher than that of Trolox. Ethanolic extraction of the dried solids yielded 7% non‐volatile solubles, containing 5% phenolics and 1.2% fucoxanthin. After 12 months at room temperature in the dark, phenolic and fucoxanthin content, and antioxidant properties of the extracts were markedly reduced. The aqueous extracts from MHG‐2 and the ethanolic extracts from the dried solids showed inhibition of lipogenesis in 3TL1 cells when used at 100 µg mL−1. CONCLUSION MHG processing can be proposed both for the extraction of an aqueous fraction and for the conditioning of seaweed biomass before conventional solvent extraction of bioactives. © 2018 Society of Chemical Industry
