Four xanthones isolated from the roots of Andrographis paniculata were tested in vitro for antiprotozoal activity against Trypanosoma brucei brucei, Trypanosoma cruzi and Leishmania infantum. Compound TDR13011 (1,2-dihydroxy-6,8-dimethoxy-xanthone) showed good activity against T. b. brucei and L. infantum with a 50% inhibitory concentration (IC 50 ) of 4.6 μ μ μ μ μg/ml and 8 μ μ μ μ μg/ml respectively. Xanthones from the roots of Andrographis paniculata exhibited promising anti-protozoal activity and these compounds could be chemically modified to obtain a more potent product.

A large number of hydroxylated 2,3-diaryl-9H-xanthen-9-ones have been synthesised by two different approaches, starting either from 3-bromo-2-methyl-4H-chromen-4-one or from (E)-3-bromo-2-styryl-4H-chromen-4-ones. The former method involves Heck reactions between 3-bromo-2-methyl-4H-chromen-4-one and styrenes, leading to (E)-2-methyl-3-styryl-4H-chromen-4-ones; these condensed with benzaldehyde to give (E,E)-2,3-distyryl-4H-chromen-4-ones, which led to the desired 2,3-diaryl-9H-xanthen-9-ones under reflux in 1,2,4-trichlorobenzene. 3-Bromo-2-styryl-4H-chromen-4-ones were obtained either by aldol condensations between 3-bromo-2-methyl-4H-chromen-4-one and benzaldehydes, or through Baker-Venkataraman rearrangements of 2-acetylphenyl cinnamates, followed by one-pot bromination/ cyclisation with phenyltrimethylammonium tribromide. A new bis-xanthone (xanthone ¼ 9H-xanthen-9-one), named bigarcinenone A (1) which is the first example of a bis-xanthone with the xanthone -xanthone linkage between an aromatic C-atom and a C 5 side chain from a guttiferae plant, a new phloroglucinol (¼ benzene-1,3,5-triol) derivative, named garcinenone F (2), together with seven known xanthones were isolated from the bark of Garcinia xanthochymus. Their structures were elucidated by spectroscopic methods, especially 2D-NMR techniques. Bigarcinenone A (1) exhibited potent antioxidant activity in the 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging test with a IC 50 value of 9.2 mm, compared to the positive control, the well-known antioxidant butylated hydroxytoluene (BHT) with a IC 50 of 20 mm ( Table 3).
