However, there is a lack of experimental evidence which supports the theory that these peptides can exert an effective antimicrobial effect. The aim of this study was to investigate the anti-listerial activity of such peptides in Asiago d'Allevo cheese. The water-soluble fraction of Asiago was ultrafiltrated through 10 kDa cutoff membranes before being dialyzed (100-500 Da cut-off) to remove proteins, salts, and organic acids, prior to freeze-drying. The growth of Listeria innocua LRGIA 01 and of Listeria monocytogenes strain 162 in Brain Heart Infusion (BHI) at 30°C in the presence or absence of 5 to 40 mg⋅mL −1 of cheese water-soluble extracts (WSEs) lyophilisate was monitored spectrophotometrically. WSEs lyophilisates from cheeses of different production systems (total mixed-ration-fed cows and cheese-making in a dairy plant in May; alpine-grazing cows and cheese-making in situ in July and September) and ripened for 6, 12, and 18 months were tested. A limited dosedependent inhibition of growth was observed at all the assayed concentrations. The addition of cheese WSEs lyophilisates to BHI broth inhibited the growth of <50% of L. innocua LRGIA 01 and <10% of L. monocytogenes strain 162. Antilisterial activity was only significantly affected by ripening time (it was maximal at 6 months). Antimicrobial textiles (65% cotton – 35% polyester) were functionalized using a patented technology that combines an antimicrobial molecule – polyhexamethylene biguanide (PHMB) and a precipitating agent – sodium laurylsulphate. Surface characterization was performed by x‐ray photoelectron spectroscopy and time‐of‐flight secondary ion mass spectrometry, and both techniques made clear signatures of PHMB at the top surface of treated textiles. Washing led to a strong decrease of PHMB concentration at the surface. Comparison of textile surface analysis and antimicrobial tests indicated that the amount of PHMB at its extreme surface of textiles after five or 10 industrial washings was sufficient to inhibit Listeria innocua but not Pseudomonas aeruginosa growth. The viability of L. innocua cells after contact with PHMB‐treated textile after one industrial washing was estimated using the Live/Dead BacLight kit (Molecular Probes, Eugene, OR): the combination of epifluorescence microscopy observations coupled with classic enumeration allowed detection of the presence of viable but nonculturable cells. PRACTICAL APPLICATIONS Protective clothing is required in the food‐processing industry to protect products from being contaminated by microorganisms carried by workers' clothes or filtration systems. Consequently, there is an increasing interest in the use of antimicrobial functionalized textiles in the food industry to avoid that textiles could be vectors for pathogenic or food spoilage microorganisms. In the present study, the correlation between PHMB (the antimicrobial agent) at the surface of textiles (monitored by surface analysis characterization methods) and their antibacterial activity was assessed. After contact with antimicrobial textiles, the enumeration of bacteria was performed either by plate counting or by direct observation by epifluorescence microscopy in the presence of fluorescent viability markers in order to determine whether viable but nonculturable bacterial cells were present.

Silk fibroins have good biocompatibility and could be used to form a variety of regenerated functional biomaterials. In this study, enzymatic oxidization of silk fibroins with tyrosinase (TYR) was carried out, followed by coupling of ε-polylysine (ε-PLL) for improving antibacterial ability of the fibroin-based biomaterial. Trinitrobenzene sulfonic acid (TNBS) was selectively used to incubate with silk fibroins prior to TYR treatment, aiming at preventing the self-crosslinking of silk fibroins during enzymatic oxidation. The results indicated that tyrosine residues in silk fibroins could be converted to reactive dioxyphenylalanine and o-quinone residues TYR successively. TNBS pretreatment inhibited the self-crosslinks of silk fibroins and promoted the successive coupling of ε-PLL to fibroin proteins with high graft yield. The combined use of TNBS, TYR, and ε-PLL treatments endowed fibroin membrane with satisfactory antibacterial ability against Staphylococcus aureus, and the obtained durability was also acceptable. The changes in surface potential and amine acid composition for the fibroin membranes verified the favorable actions of the combined treatment. The present method could be potentially utilized for enzymatic functionalization of various fibroin-based biomaterials.
