Abnormal expression of pro-inflammatory mediators such as cell adhesion molecules and cytokines has been implicated in various inflammatory skin diseases, including atopic dermatitis. In this study, we investigated the anti-inflammatory activity of Aronia melanocarpa concentrate (AC) and its action mechanisms using in vivo and in vitro skin inflammation models. Topical application of AC on mouse ears significantly suppressed 12-O-tetradecanoylphorbol-13-acetate (TPA)-induced ear edema formation, as judged by measuring ear thickness and weight, and histological analysis. Topical administration of AC also reduced the expression of pro-inflammatory cytokines such as TNF-α, IL-1β, and IL-6 in TPA-stimulated mouse ears. Pretreatment with AC suppressed TNF-α-induced ICAM-I expression and subsequent monocyte adhesiveness in human keratinocyte cell line HaCaT. In addition, AC significantly decreased intracellular reactive oxygen species (ROS) generation as well as mitogen-activated protein kinase (MAPK) activation in TNF-α-stimulated HaCaT cells. AC and its constituent cyanidin 3-glucoside also attenuated TNF-α-induced IKK activation, IκB degradation, p65 phosphorylation/nuclear translocation, and p65 DNA binding activity in HaCaT cells. Overall, our results indicate that AC exerts anti-inflammatory activities by inhibiting expression of pro-inflammatory mediators in vitro and in vivo possibly through suppression of ROS-MAPK-NF-κB signaling pathways. Therefore, AC may be developed as a therapeutic agent to treat various inflammatory skin diseases.

To get a more comprehensive knowledge of oil contents and fatty acid pattern, seed oils from various Rosaceous plants belonging to the subfamilies Maloideae and Rosoideae, respectively, were investigated. For this purpose, isolated seeds of 18 dessert and cider apple (Malus domestica BORKH.) cultivars of different provenances, pear (Pyrus communis L.), rose hip (Rosa canina L.), quince (Cydonia oblonga Mill.), and red chokeberry (Aronia arbutifolia L.) were analyzed for their oil content and fatty acid composition. Oil contents varied significantly, not only among the different genera, but also among cultivars of one species, ranging from 0.8 to 29.4 g/100 g dry matter. Qualitatively, the fatty acid profiles of the investigated seed oils showed good agreement in all representatives of the Rosaceae. Their triacylglycerols were uniformly composed of linoleic, oleic, palmitic, stearic, palmitoleic, a-linolenic, arachidic, gondoic, and behenic acids. Quantitation of individual fatty acids revealed the oils to be rich in mono-and diunsaturated oleic acid and linoleic acid, ranging from 15.1 to 33.3 g/100 g and from 32.5 to 49.7 g/100 g, respectively. As expected, contents of saturated fatty acids were 6-10 times lower. Moreover, apple cultivars showed pronounced differences in yields, numbers, and weights of their seeds. As demonstrated by the data obtained from this study, seeds resulting from the processing of apple, pear, quince, chokeberry (Maloideae), and rose hip (Rosoideae) into juices, jellies, and jams may serve as a promising source for the recovery of nutritionally valuable edible oils.

OBJECTIVEThis study investigated the effects of resveratrol, a natural polyphenol with neuroprotective properties, on retinal neuronal cell death mediated by diabetes-induced activation of Ca2+/calmodulin-dependent protein kinase II (CaMKII).RESEARCH DESIGN AND METHODSDiabetes was induced in C57BL/6 mice by five consecutive intraperitoneal injections of 55 mg/kg streptozotocin (STZ). Control mice received buffer. All mice were killed 2 months after the injections, and the extent of neuronal cell death, CaMKII, and phospho-CaMKII protein expression levels and CaMKII kinase activity were examined in the retinas. To assess the role of CaMKII in the death of retinal neurons, a small-interfering RNA (siRNA) or specific inhibitor of CaMKII was injected into the right vitreous humor, and vehicle only was injected into the left vitreous humor, 2 days before death. Resveratrol (20 mg/kg) was administered by oral gavage daily for 4 weeks, beginning 1 month after the fifth injection of either STZ or buffer.RESULTSThe death of retinal ganglion cells (RGCs), CaMKII, phospho-CaMKII protein levels, and CaMKII activity were all greatly increased in the retinas of diabetic mice compared with controls, 2 months after induction of diabetes. Terminal deoxynucleotidyl transferase (TdT)-mediated dUTP nick-end labeling (TUNEL)-positive signals co-localized with CaMKII- and phospho-CaMKII immunoreactive RGCs. However, in addition to CaMKII knockdown and inhibition by siRNA or a specific inhibitor, respectively, resveratrol provided complete protection from diabetes-induced retinal cell death.CONCLUSIONSIn the present study, resveratrol prevented diabetes-induced RGC death via CaMKII downregulation, implying that resveratrol may have potential therapeutic applications for prevention of diabetes-induced visual dysfunction.
