Purpose: To investigate the effect of Quercetin (Que) on inflammatory reaction in intestinal porcine enterocyte cells, IPEC-J2, induced by lipopolysaccharide (LPS). Methods: IPEC-J2 cells were pretreated with Que and then incubated with LPS. Cell viability, cell morphology, nitric oxide (NO) content, gene expression levels of interleukin-6 (IL-6) and interleukin-8 (IL-8), were then evaluated. Results: LPS incubation at 10 μg/mL for 24 h showed no effect on the viability and cell morphology of IPEC-J2, while Que pre-incubation significantly enhanced the viability of IPEC-J2 and improved cell morphology (p < 0.05). Que decreased the expression levels of pro-inflammatory cytokines IL-6 mRNA and IL-8 mRNA raised by LPS (p < 0.05). Interestingly, both LPS and Que demonstrated no influence on the release of NO. Conclusion: Pre-treatment of Que shows a positive effect on intestinal porcine enterocyte cells and inhibited porcine intestinal inflammation in vitro induced by LPS, but the mechanism may be not associated with NO-related signaling pathway. Therefore, Que might have a potential effect as a veterinary drug or feed additive for the treatment of enteritis in pigs.

Korean mistletoe has anti-inflammatory and antioxidant functions and may be a useful training supplement. We investigated the effect of Korean mistletoe extract (KME) on inflammatory markers after high-intensity exercise by 20 university male rowers (KME group vs. CON group) consuming 110 mL KME/dose (2 times a day over 8 weeks). Blood samples were collected for measurement of serum cytokine levels at baseline, immediately after exercise, and following 30 minutes of recovery. Interleukin-6 (IL-6), tumor necrosis factor-alpha (TNF-α), and C-reactive protein (CRP) were used as markers for inflammation. After supplementation, IL-6 and TNF-α levels were significantly lowered in the KME group than in the CON group at baseline, immediately after exercise, and following 30 minutes of recovery. KME can reduce high-strength exercise-induced increases in the levels of serum inflammatory cytokines in active individuals and improve anti-inflammatory functions.

Hydroxyurea (HU), a DNA synthesis inhibitor, is one of the most common chemotherapeutic drugs that have been widely applied to treat a variety of cancers. HU treatment exhibits severe side effects including renal toxicity, skin toxicity and embryo-toxicity. However, the influence of HU on the female gamete development has not yet fully clarified. Here, we found that HU exposure induced the degeneration of activated follicles after primordial follicle stage, resulting in the depletion of the ovarian reserve. HU exposure also led to the oocyte meiotic maturation arrest via disrupting normal spindle assembly, chromosome alignment and kinetochoremicrotubule attachment. Furthermore, exposure to HU impaired the dynamics of ovastacin and Juno, two critical fertilization regulators. Notably, we illustrated that Shoutai pills (STP), a traditional Chinese medicine drug that has been commonly used for the treatment of miscarriage in China, partially restored all of the defects of oocyte development resulting from HU exposure through inhibiting the occurrence of oxidative stress-induced apoptosis. Taken together, our data not only reveal the adverse impact of HU exposure on the female gamete development, but also provide an effective strategy to prevent it, potentially contributing to the improvement of the quality of oocytes from patients treated with HU.
