Karkatshringi (Pistacia integerrima) is an essential Ayurvedic medicinal plant belonging to the family Anacardiaceae. The plant originated in India and grows in the ranges of the north-western Himalayas. It is distributed over China, Afghanistan, Pakistan, and Nepal. The flowers of this plant is small and red, fruits are shiny and brown. Galls are hollow, horn-shaped, bitter and used for medicinal purposes. Gall powder is most commonly used in diseases such as cough, diarrhoea, fever, etc. also effective against staphylococci, pseudomonas and Escherichia coli bacteria. The different parts of the plant contain amino acids, sterols, alkaloids, flavonoids, tannins and saponin. In Pistacia integerrima, the volatile chemical constituents are extracted by hydro distillation or liquid-liquid extraction. Pistacia integerrimais associated with fungi which causes rust and blight diseases. Traditionally it is used for rheumatic pain, analgesic, and antipyretic effects. Karkatshringi is an important ingredient of Ayurvedic formulations like Chavyanprash, Dashmularista and Shringyadi Churna. Galls are used in the treatment of diarrhoea. Diabetic patients also use it. Essential oil of Pistacia integerrima was used for in-vitro studies like antioxidant activity, angiogenesis, and mast cell degranulation. Karkatshringi studied for its various activities such as anti-inflammatory, anti-oxidant, antibacterial, anti-cancer, anti-diarrhoeal, anti-convulsant, muscle relaxant, etc. This article includes detail information of Pistacia integerrima related to chemical constituents and pharmacological activities. INTRODUCTION:As per Ayurveda, karkatshringi is the most important medicinal plant. The galls of Pistacia integerrima are typically used in the treatment of paediatric disease. According to Acharya Charaka, the plant consists of the Hikkanigrahan and Kasaharagana, which are explained in vatajKasa and Kapha 1 . As a moderate-sized, broadleaf tree, the karkatshringi can be found in Nepal, China, Afghanistan, Pakistan and the northwest& West Himalayas ranges 2 .

Dioscorea bulbifera is commonly known as Aerial Yam is a perennial, non-spiny, climbing plant consist of numerous medicinal benefits. It contains a large number of polyphenols including organic acids and saponins. Aim of the present study is to screen the anti-inflammatory activity of saponin from the aqueous methanol fruit extract of Dioscorea bulbifera. The presence of saponin was tested using the qualitative phytochemical screening and separated using TLC. Total saponin content was 30µg/g. The identification of saponin activity against inflammation was carried out using several assays such as Cyclooxygenase inhibition assay, Lipooxygenase Inhibition assay and Protein denaturation inhibition assay using Diclofenac sodium. In silico study was conducted for the evaluation of anti-inflammatory activity using the COX-2 as target. The docking results revealed that dioscin was predicted to have the highest binding efficacy with COX-2 receptor with a binding energy of -662.07 kcal/mol. Thus the study shows that the Dioscorea bulbifera fruit contain a safe and effective Anti-inflammatory activity.

Monosodium iodoacetate induces Cartilage degradation and inflammation in rats in a dose- and time-dependent manner

Monosodium iodoacetate (MIA) is a widely recognized agent for inducing cartilage degradation and inflammation in animal models. In this study, we systematically investigated the dose- and time-dependent effects of MIA on cartilage degradation in the knees of rats. We evaluated knee diameter, knee bend score, and the levels of pro-inflammatory cytokines (IL-1 and IL-8) and cartilage degradation markers (CTX-II) to gain insights into cartilage damage and inflammation progression. Wistar rats were categorized into groups receiving various doses of MIA via intra-articular injection (0.1 mg, 0.5 mg, 1 mg, 3 mg, and 5 mg) and were observed at multiple time intervals (2, 4, 6, 8, 10, and 12 weeks). We measured knee diameter to gauge joint swelling, assigned knee bend scores to assess functional limitations, and analyzed synovial fluid samples for IL-1β, IL-8, and CTX-II levels. Our data show that low-dose MIA increases pain sensitivity, knee diameter, inflammatory cytokines IL-1β and IL-8, and CTX-II levels, which progress slowly over 12 weeks. On the other hand, higher dosages of MIA caused cartilage deterioration after two weeks, followed by an increase in inflammatory cytokines. This research sheds light on MIA administration's dose- and time-dependent effects on cartilage breakdown and inflammation in rat knee joints. Analyzing knee diameter, knee bend score, IL-1β, IL-8, and CTX-II as evaluation parameters provides a multidimensional perspective of cartilage injury and inflammatory dynamics. These findings help us comprehend cartilage-related illnesses and have implications for future research on therapeutic approaches for cartilage disorders.
