ResumenLos radicales libres y el daño oxidativo están relacionados con la muerte celular, siendo la peroxidación lipídica un mecanismo que lleva a la destrucción oxidativa de la membrana celular. Además, en procesos inflamatorios, uno de los primeros estadios es la permeabilidad vascular incrementada (formación del edema) seguido de la extravasación de los fluídos, migración de leucocitos al sitio dañado y liberación de proteínas pro-inflamatorias. Evaluamos el efecto antioxidante y antiinflamatorio para Petiveria alliacea "mucura". Para el efecto antioxidante, se evaluó la formación de especies reactivas al ácido tiobarbitúrico como indicador de la peroxidación lipídica. Dosis 200mg/mL de Petiveria alliacea disminuyó significativamente en un 42% los niveles de MDA comparado con el agua (control negativo). En la evaluación antiinflamatoria, se indujo la inflamación por inyección de carragenina (solución al 1%), en la parte subplantar de ratones y en la "bolsa de aire subcutánea" de ratas para la inflamación aguda y crónica respectivamente. En la evaluación antiinflamatoria hay una máxima reducción del edema en un 23,26% a las 4 horas del tratamiento. Para la inflamación crónica hay una reducción del 25,9% y 29,5% del peso y volumen del exudado extraído, respectivamente, así como una reducción del 24% de peso de tejido fibroso. Estos resultados evidencian efecto antioxidante y antiinflamatorio de Petiveria alliacea.Palabras clave: Petiveria alliacea; antioxidante; antiinflamatorio; peroxidación; radical libre. AbstractThe free radicals and the oxidative damage are related with cellular death, with the lipid peroxidation being a mechanism that leads to oxidative destruction of the cellular membrane. Furthermore, in inflammatory process, one of the first stages is the incremented vascular permeability (formation of the oedema) followed with the extravasation of the fluids, migration of the leucocytes to the damaged site and release of pro-inflammatory proteins. Was evaluated antioxidants and anti-inflammatory of Petiveria alliacea "mucura". For antioxidants effect, was evaluated formation of thiobarituric acid reactive substances as indicator of lipid peroxidation. Doses 200 mg/mL of Petiveria alliacea decrease significatly in 42% levels of MDA compared to water (negative control). In the antiinflammatory evaluation, inflammation was analyzed in the carrageenan-induced paw oedema test in mice (solution to 1 %) and in the "carrageenan-induced air-pouch formation" of rates for acute and chronic inflammation respectively. In anti-inflammatory evaluation there is a significant reduction of the edema in 23.26% at 4hrs at the treatment. For chronic inflammation there is a reduction of 25.9% and 29.5% of the weight and volume of exuded extract respectively, as well as a reduction of 24% of weight of fibrous tissue. Context: Perovskia atriplicifolia Benth (Labiantae) has long been used as a traditional herbal medicine for anti-inflammation in Pakistan; this prompted us to isolate anti-inflammatory compounds from this plant. Objective: The objective of this study was to isolate and characterize the anti-inflammatory principles from Perovskia atriplicifolia. Materials and methods: The CHCl 3 -soluble fraction of the methanol extract of the whole plant on column chromatography yielded compounds 1-6. The anti-inflammatory potential of the compounds 1-6 was evaluated by Leukotriene C4 (LTC 4 ) Release Assay which was performed according to the established protocol. LTC 4 in the supernatant of each well was measured using an ELISA kit (Cayman Chemical Co., Ann Arbor, MI). Results: The bioassay-guided phytochemical investigation of the CHCl 3 soluble fraction of the methanol extract of Perovskia atriplicifolia furnished six compounds, abrotanone (1), abrotandiol (2), (+)-pinoresinol (3), (+)-syringaresinol (4), (+)-lariciresinol (5), and (+)-taxiresinol (6). The compounds (1-6) were evaluated for their inhibitory activities on LTC 4 release. Among the tested compounds, (+)-taxiresinol (6) exhibited the most potent inhibition of LTC 4 release with an IC 50 value of 3.4 ± 0.09 mM followed by compounds 4, 5, 3, and 2 with an IC 50 value ranging from 7.9 ± 0.04 to 17.2 ± 0.07 mM. Abrotanone (1) showed the lowest inhibition of LTC 4 release with an IC 50 value of 35.1 ± 0.05 mM (the positive control, zileuton, 0.77 ± 0.05 mM). Conclusion: Compounds 1-6 were found to possess inhibitory activity and seem to have potential therapeutic effect on inflammatory diseases.

Euphorbia antiquorum (L.) is used as traditional medicine for various ailments in Bangladesh. But the scientific basis for its use especially in pain and inflammation remains largely unknown. Therefore, the present study was designed to evaluate analgesic and anti-inflammatory effect of the aqueous ethanolic extract of the whole plant. The analgesic activity was evaluated by hot plate, acetic acid induced writhing and formalin induced writhing methods in Swiss Albino mice at the doses of 250 and 500 mg kg −1 body weight. The extract was also investigated for the anti-inflammatory effect on Long Evans rats at above mentioned doses using carrageenan induced rat paw edema method. Phytochemical analysis of the extract revealed the presence of tannins, alkaloids, flavonoids, saponins and terpenoids. The extract elicited a significant (p<0.01) analgesic activity in a dose dependent fashion in both the early and late phases of formalin test and also in hot plate and acetic acid induced writhing tests at both the doses employed. In acetic acid induced writhing test, the extract (at 500 mg kg −1 ) produced a maximum inhibition of writhing reaction by 51.13% (p<0.001), which is comparable to that of the standard drug Diclofenac sodium (58.80%). The extract also significantly inhibited the licking response at the dose of 500 mg kg −1 in both the early phase (63.64%, p<0.01) and the late phase (66.43%, p<0.01) of formalin test while the standard drug inhibited by 64.20% and 72.03%, respectively. The oral administration of the extract significantly (p<0.001) inhibited inflammatory response induced by carrageenan in a dose dependent manner. The most prominent inhibition of 74.25% (250 mg kg −1 ) and 78.44% (500 mg kg −1 ) were observed at the 4th hour of study. Presence of various chemical constituents in the extract of Euphorbia antiquorum especially tannins, flavanoids, terpenoids and alkaloids might be responsible for the observed analgesic and anti-inflammatory effects. These experimental findings lend pharmacological support to the suggested folkloric uses of the plant in the management and/or control of pain as well as inflammatory conditions.
