Anti‐Hyperglycemic Properties of Crude Extract and Triterpenes from Poria cocos

Abstract: Poria cocos, Bai Fu Ling in Chinese, is used in traditional Chinese medicine to treat diabetes. However, its claimed benefits and mechanism are not fully understood. This study aimed to investigate the effect and action of P. cocos on type 2 diabetes. We first performed phytochemical analysis on the crude extract and factions of P. cocos. P. cocos crude extract at 50 mg/kg body weight or more significantly decreased blood glucose levels in db/db mice. Based on a bioactivity-directed fractionation and isolation… Show more

“…Ion detection was performed in multiple reaction monitoring (MRM) mode wherein the m/z 527.4 527. 4 We assessed potential analyte interference in the LC peak region with six different blank rat plasma samples using the proposed extraction procedure and LC-MS/MS conditions. Quality control (QC) stock solutions of PA (1 mg/mL) and GA (1 mg/mL) were prepared by dissolving the standards in acetonitrile, respectively.…”

“…1) P. cocos alone or in combination with other herbs is often used to treat diabetes as well as other disorders. [2][3][4] The main ingredients of this herbal medicine include a group of triterpenoid compounds. Pachymic acid (PA) is one of the major components in P. cocos and plays an important role in various pharmacological effects.…”

“…Similarly, dehydrotrametenolic acid from Poria cocos Wolf, a mushroom used in traditional Chinese medicine to treat diabetes, was suggested to act as an insulin sensitiser through the action of PPARγ [68]. However, because it does not activate the PPARγ pathway, the enhanced insulin sensitivity induced by dehydrotrametenolic acid has been suggested to be irrespective of PPARγ [69]. It is important to remark that the lack of adipogenic activity and/or transcriptional activity of PPARγ in the presence of a compound must not be considered an evidence that their antidiabetic activity is not mediated by PPARγ.…”
