Anti-HIV Cyclotides

Abstract: The cyclotides are a recently discovered, structurally unique family of bioactive plant peptides. Their discovery spawned a series of structural analyses, synthetic efforts, and studies to define the biosynthesis and biological properties of these novel peptide metabolites. Cyclotides have a head-to-tail cyclized amino acid backbone and a conserved cystine knot motif that provides an extremely stable structural framework. They all share a common global fold and are highly resistant to denaturation and to cleav… Show more

“…The plant has a strong folk reputation in the treatment of cancer. Viola has been investigated for cytotoxicity and reported as a pharmacological tool serving as antitumor agents (Lindholm et al 2002;Mokhtari 2016;Gustafson 2004;Ireland et al 2006;Ebrahimzadeh et al 2010). …”

“…Cyclotides 1,2 are a unique class of head-to-tail cyclic cysteine-15 rich microproteins up to 37 amino acids in length that exhibit a wide range of biological properties, ranging from anti-HIV [3][4][5] to insecticidal activity. 6,7 In contrast to many smaller naturally occurring cyclic peptides they are true gene products, and possess a highly stable cystine-knot † topology, 20 whereby two disulfide bridges form a ring through which a third is threaded.…”

“…The cyclotides are a distinctive class of plant defense peptides produced by various species in the Violaceae and Rubiaceae Craik, 2001;Gustafson et al, 2004;. They are characterized by their head-to-tail cyclization and cystine knot motif , which contribute to their exceptional resistance to physical, chemical, and biochemical degradation (Gran, 1973a;Colgrave and Craik, 2004).…”
