Anti-Endotoxin Monoclonal Antibodies Inhibit Secretion of Tumor Necrosis Factor-a by Two Distinct Mechanisms

Abstract: These results suggest that IgG and IgM anti-LPS mAbs exert protective capacity by extracellular neutralization of LPS, while IgG Fc receptor-mediated cellular uptake also may serve to bypass macrophage activation and TNF-alpha secretion by promoting internalization and intracellular neutralization.

“…They could affect the immune response of the mother mice and consequently that of the neonate by neutralization of LPS. An effect of anti‐LPS antibodies on the cytokine production profile of macrophages has been previously reported [31]. In conclusion our study provides first experimental evidence for the concept that prenatal exposure to microbial components — LPS in this study – act as potent immuno‐modulators for the development of respiratory allergies later in life.…”

“…Historically, anti-LPS mAbs have been very successful in animal models in blocking sepsis and improving survival when administered prior to a septic insult (15,(31)(32)(33). These results were so promising and sepsis has such a high mortality that two anti-LPS mAbs were developed for human use.…”

“…Because of the higher opsonic activity of IgM antibodies, one can expect that IgM anti-LPS will enhance the complement-mediated internalization of LPS by macrophages better than will IgG anti-LPS [22,35]. Fc-mediated and opsonin-mediated antibody interactions may play an important role in the protective activity of anti-LPS antibodies [10]. The lack of the IgM fraction might also explain the limited protection of i.v.…”
