Anti-DNA Investigated by the DNA Bentonite Flocculation Technique

Abstract: The DNA bentonite flocculation test was used to investigate the presence of antibody to DNA. In most sera a prozone-like phenomenon was found, caused by a non-specific inhibition of serum. Anti-DNA was found with extreme frequency in patients with SLE, chronic liver disease, acute hepatitis, infectious mononucleosis, and blood donors over the age of 60, but was found rarely or not at all in patients with some other diseases or blood donors under 60 years of age.

A characteristic feature of immunologic diseases is their association with each other. For multiple sclerosis (MS), several retrospective studies reported increased as well as expected coincidence rates with other immunologic diseases. We conducted a prospective case-control study of MS patients and healthy volunteers and found 13/101 MS patients and 2/97 controls with such diseases (P = 0.009, chi-square test), as well as 47/88 MS patients versus 31/95 controls with a variety of circulating autoantibodies (P = 0.004, chi-square test). SUMMARY In investigations by counter-immunoelectrophoresis, anti-actin antibodies were found in 59°of patients with chronic hepatitis and in 8 % of patients with non-hepatic diseases and normal blood donors. Anti-actin antibodies were found more frequently in patients with hepatitis and IgG smooth muscle antibodies than in other groups of diseases and normal subjects with IgG smooth muscle antibodies. Anti-actin antibodies showed no correlation with bile canalicular antibodies.
