Anti-angiogenic effects of non-peptide integrin αvβ3 specific antagonist on laser-induced choroidal neovascularization in mice

Abstract: The integrin alphavbeta3 may play a key role in the induction of laser-induced CNV. The antagonists for integrin alphavbeta3 may have therapeutic effects in CNV associated diseases such as age-related macular degenerations.

“…5) (51). Similar inhibition of choroidal vessel growth was previously obtained with antagonist a5b1 (16,21) and avb3 (19,20) antagonists. Surprisingly, higher inhibition of neovascular tufts was reached with EMD478761, a compound that inhibits both avb3 and avb5 integrins; however, this compound was effective only when sustained amounts were injected into the vitreous cavity by an intravitreal delivery device (17).…”

“…Cyclic peptides that mimic tripeptide Arg-Gly-Asp (RGD) binding, motif of integrin avb3, Abs directed against integrin subunits, and molecules that are isolated from venoms have been shown to disrupt integrin/basal membrane recognition and have been used in animal models of cancer, fibrosis, and inflammation (15). Their systemic or local (intravitreal) administration has been shown to inhibit CNV in a laser-induced CNV model and retinal neovascularization in the mouse oxygen-induced retinopathy (OIR) model (16)(17)(18)(19)(20)(21).…”

“…Previous reports show that integrin αvβ3 is involved in ocular angiogenesis [22], [23]. In vivo experiments have shown antibodies blocking or immunoconjugate drug therapy targeting integrin αvβ3 inhibit neovascularizaion [17], [23][26]. In addition, integrin αvβ3 potentiates the internalization of various viruses [27], [28], making it a potential target for drug delivery via liposome based nanoparticles.…”

“…16 VEGF, antibody blocking, and immunoconjugate drug therapy targeting integrin α v β 3 inhibit NV in corneal/choroidal NV. 16,44,45 The corneal epithelium and stroma express α 2 β 1 as well as α 11 β 1 , while the normal epidermis and corneal epithelium lack α 5 β 1 and α v β 3 . 46,47 Therefore, integrin α v β 3 heterodimers on vascular endothelial cells could be recognized by GEH-RGD from the interaction of RGD on the particles.…”
