Anti-Alzheimer Activity of Combinations of Cocoa with Vinpocetine or Other Nutraceuticals in Rat Model: Modulation of Wnt3/β-Catenin/GSK-3β/Nrf2/HO-1 and PERK/CHOP/Bcl-2 Pathways

et al.

Abstract: Alzheimer’s disease (AD) is a devastating illness with limited therapeutic interventions. The aim of this study is to investigate the pathophysiological mechanisms underlying AD and explore the potential neuroprotective effects of cocoa, either alone or in combination with other nutraceuticals, in an animal model of aluminum-induced AD. Rats were divided into nine groups: control, aluminum chloride (AlCl3) alone, AlCl3 with cocoa alone, AlCl3 with vinpocetine (VIN), AlCl3 with epigallocatechin-3-gallate (EGCG)… Show more

“…For instance, the nuclear factorerythroid 2-related factor 2 (Nrf2) pathway reduces OS by manipulating antioxidant proteins such as heme oxygenase-1, mitigating inflammation, and inhibiting Aβ and tau phosphorylation, which enhances memory, learning, and cognitive abilities. 34,35 Research has also reported that miR-146a induces OS through the MAPK signaling pathway, exacerbating cognitive impairment and AD pathology. 36 Inhibition of the MAPK signaling effectively ameliorates synaptic damage and neuroinflammation.…”

“…Moreover, it has been reported that after 8 weeks of supplementation with cacao powder, d-galactose-induced aging rat brains were found to have increased levels of free radical scavenging enzymes such as catalase and glutathione peroxidase ( Yoo and Kim, 2021 ). It has been reported that after 5 weeks of oral administration, the combination of cacao with nutraceuticals moderates inflammation, antioxidant responses, GSK-3-Wnt/-catenin signaling, ER stress, and apoptosis in aluminum chloride-induced AD rats ( Abu-Elfotuh et al, 2023 ). It may, therefore, be plausible to speculate that cacao’s ability to scavenge free radicals and prevent oxidative damage contributes to its protective effect against Aβ-induced hippocampal LTP impairment.…”
