Anthrone in Estimating Low Concentrations of Sucrose

Abstract: A procedure is presented for the estimation of sucrose concentrations in the range of 10 to 250 parts per million, using a 0.05% solution of anthrone in concentrated sulfuric acid. This method offers the advantages of increased sensitivity and accuracy over the commonly used Molisch -naphthol test.

“…Soluble carbohydrates were extracted by treating replicate (n = 2) subsamples of 200 mg with hot (100°C) 2 M HCl for 2.5 h (Hashimoto et al 1987). Total soluble carbohydrates were determined by the Anthron method (Morse 1949) using glucose as a standard.…”

“…Reducing sugar content of chickpea seeds was estimated by DNS (Dinitrosalicylic acid) method (Miller 1972). The starch content of chickpea seeds was determined by the anthrone method (Morse 1947, Jayaraman 1985 and lipid content of chickpea seeds was determined by the method of Bligh and Dyer (1959).…”

“…Samples of lung homogenate supernatants were added to a solution of 0.05% anthrone (Sigma) in sulfuric acid, mixed well, and allowed to equilibrate for 20 min. Ficoll concentrations were determined by measuring absorbance spectrophotometrically at 627 nm and represent the total Ficoll retained per lung (16).…”
