Anthrax Lethal Toxin Inhibits Translation of Hypoxia-inducible Factor 1α and Causes Decreased Tolerance to Hypoxic Stress

Abstract: Background: Hypoxia is proposed as a mediator of anthrax lethal factor (LT)-induced pathology. Results: LT inhibits hypoxia-inducible factor 1␣ (HIF-1␣) translation and causes increased cellular toxicity in response to hypoxic stress. Conclusion: LT reduces HIF-1␣ translation, dysregulating host responses to hypoxia. Significance: Inhibition of HIF-1␣ translation is a novel mechanism underlying LT-induced pathology.

“…LF is a zinc-dependent metalloprotease with specific activity against MKKs. Consistent with our previous studies (22,24), LT treatment led to cleavage and degradation of MKK1, MKK2, MKK3, MKK4, and MKK6, but not MKK7, in Hepa1c1c7 cells (Fig. 4A).…”

“…We have reported previously that LT inhibits the accumulation of hypoxia-inducible factor (HIF) 1␣, the master regulator of cellular responses to hypoxia, by blocking its translation (22). To investigate whether LT inhibits c-Jun by a similar mechanism, we first investigated the effect of LT treatment on de novo c-Jun protein synthesis.…”

“…The changes in active translational factors causes decreased translation of Hypoxia-Induced Factor-1a (HIF1 a), a central protein involved in the host cell response against hypoxic condition. Severely decreased expression of HIF-1a could lead to pathological conditions in host like angiogenesis and metastasis [50]. The implications of N-terminus cleavage of MAPKK family members have been found to be diverse.…”
