Antenatal Dexamethasone Administration Increases Fetal Lung DNA Synthesis and RNA and Protein Content in Nitrofen-Induced Congenital Diaphragmatic Hernia in Rats

Abstract: Antenatal glucocorticoids treatment has been shown to correct pulmonary immaturity. The thymidine analog bromodeoxyuridine (BrdU) is incorporated into S-phase cells and used as a marker of DNA synthesis. In this study, we investigated the effect of antenatal glucocorticoid administration on DNA synthesis and RNA and protein content in nitrofen-induced congenital diaphragmatic hernia (CDH) in rats to better understand the effect of antenatal glucocorticoids on CDH lung. The CDH model was induced in pregnant rat… Show more

“…Regarded to the histological aspects of the liver and kidneys the results did not show changes in the structural level of the neonates from group I. These findings are according to the literature that points out for the existence of gestational periods of higher susceptibility to glucocorticoids that would be related to the variation in the level of the enzymes 11b-HSD type 1 and 2 during the gestation (Laborde et al,1992;Oue et al, 2000;Nevagi & Kaliwal, 2001;Seckl).…”

“…In the animal model of CDH, corticosteroids have been used to try promoting lung growth in uterus [42,43]. However, a recent clinical trial on the prenatal use of corticosteroids in human CDH failed to demonstrate any benefits [6].…”

“…Its expression during embryogenesis had been studied in details in the mouse. Expressing at normal level, it plays a central role in lung branching, differentiation of the primordial ducts into alveolar, and generation of the bronchiolar ducts [23]. Several studies have examined the effect of abnormal amounts of TGF-β1 on lung development.…”
