Background Entomopathogenic fungi exists naturally in plants as an asymptote and have the potential to reduce the population of insect pests through indirect interactions. This study was conducted to detect and characterize the endophytic fungi Beauveria bassiana and Metarhizium robertsii from the rhizosphere soil of tomato plants and their virulence effect on Galleria melonella. Methods From the rhizosphere soil of 40 tomato fields, three Beauveria bassiana and seven Metarhizium robertsii isolates were isolated using the galleria bait method. All fungi isolate were morphologically characterized by their colony color, shape, and surface texture. Isolates with the highest percentages of germination, conidial yield, and radial growth were selected, and their virulence was evaluated on second instar larvae of Galleria melonella under laboratory conditions. Results In this study, Beauveria bassiana showed white colony color and aseptate hyphae, whereas Metarhizium robertsii showed dark green to light green colony color and septate hyphal structures. Maximum spore production and conidial length were obtained by Beauveria bassiana isolate APPRC-27 with 2.67x107 spores ml-1 and 3.24 µm, respectively. Colony radial growth rates ranged from 1.73 to 3.24 mm day-1. The results revealed that the highest mortality rate of Galleria melonella (100%) was obtained by Metarhizium robertsii isolates K-61 and K-102 at a concentration of 1x108 conidial ml-1 at 7 days post-inoculation. The lowest mortality rate was registered by Metarhizium robertsii isolate RST-11. Conclusions In the present study, isolates that produced the most spores and had the highest germination rates were the most virulent to Galleria mellonella second instar larvae. Therefore, virulent entomopathogenic fungi, Beauveria bassiana and Metarhizium robertsii, are promising bioagents for the control of insect pests.

Fusarium oxysporum f. sp. cubense merupakan cendawan tular tanah penyebab penyakit panama pada tanaman pisang. Agens hayati dari kelompok cendawan telah banyak dilakukan untuk mengendalikan penyakit tanaman. Oleh sebab itu, penelitian ini bertujuan mengidentifikasi empat koleksi cendawan entomopatogen berdasarkan pada ciri morfologinya dan mengevaluasi mekanisme antagonismenya terhadap F. oxysporum f. sp. cubense IPBCC 19 1472. Galur cendawan entomopatogen dengan kode PS 4, PS 9, PS 11 berasal dari kawasan Situ Gunung, Sukabumi, Jawa Barat dan galur KRC berasal dari Kebun Raya Cibodas, Jawa Barat. Identifikasi cendawan dilakukan berdasarkan ciri morfologi pada medium agar-agar dekstrosa kentang. Mekanisme antagonistik diteliti menggunakan metode biakan ganda dan sebagai kontrol digunakan biakan tunggal F. oxysporum f. sp. cubense. Pengamatan dilakukan terhadap daya hambat cendawan entomopatogen dan pertumbuhan koloni F. oxysporum f. sp. cubense. Semua cendawan entomopatogen tidak dapat diidentifikasi secara morfologi karena tidak bersporulasi. Pertumbuhan koloninya lebih lambat daripada F. oxysporum f. sp. cubense. Semua cendawan entomopatogen menghambat F. oxysporum f. sp. cubense melalui mekanisme kompetisi ruang. Galur KRC memiliki aktivitas antagonisme paling besar, diikuti berturut-turut oleh galur PS 9, PS 11, dan PS 4. F. oxysporum f. sp. cubense membentuk klamidospora sebagai respons terhadap cendawan entomopatogen galur PS9 dan vakuolisasi ketika berinteraksi dengan tiga galur cendawan entomopatogen lainnya.

Hypera postica. Ciclul de viață al speciei H. postica poate fi caracterizat prin perioade scurte de activitate intensă și perioade lungi de inactivitate. Hypera postica prezintă o remarcabilă plasticitate comportamentală și ecologică (Hsiao, 1993). Cele mai multe populații sunt univoltine (Helgesen, Cooley, 1976), dar în unele medii, în special în regiunile din sud, poate fi o a doua generație (Michelbacher, 1943;White et al., 1969). Insecta iernează în stadiul de adult. În localitățile cu ierni blânde, ouăle, larvele și pupele, de asemenea, pot supraviețui iernii Radcliff e, Flanders, 1998).În funcție de regiune, în special în cele sudice, depunerea pontei poate avea loc toamna, iarna și primăvara (Woodside, Bishop, Pienkowski, 1968;Campbell et al., 1975;Whitford, Quisenberry, 1990), ori de câte ori temperaturile depășesc 1,7°C (Hsieh, Roberts, Armbrust, 1974). Tulpina tânără a lucernei este locul preferat pentru depunerea pontei (Pass, 1967), cu toate acestea, ouăle sunt depuse și în tulpinile buruienilor, printre care, Poa bulbosa L., Stellaria sp. (L.), Capsella bursa -pastoris (L.) (Ben Saad, Bishop, 1969;Niemczyk, Flessel, 1970). Odată ce locul pentru depunerea pontei este ales, femela perforează peretele plantei, introduce ovipozitorul și depune un grup de 9-10 ouă de culoare galbenă (Niemczyk, Flessel, 1970).În condiții optime de laborator, potențialul de reproducere este ≈ 3650 de ouă per femelă (Coles, Day, 1977). Rata de ovipoziție crește liniar cu temperatura până la 30°C (LeCato, Pienkowski, 1972;Hsieh, Roberts, Armbrust, 1974). Incubația necesită 156 GZ (grade zile), cu un prag al temperaturii de 9,0°C per zi (Roberts, DeWitt, Armbrust 1970). Larva apărută se alimentează cu măduva tulpinii, apoi creează galerii spre exterior. Ieșind, urcă pe mugurii foliari terminali cu care se hrănește în continuare. Insecta posedă patru stadii larvare. Temperatura limită de dezvoltare este de 8,9°C, cu suma temperaturilor necesare pentru a fi naliza dezvoltarea larvelor egală cu 148°C (Litsinger, Apple, 1973). Insecta imatură se transformă în pupă și imago, după acumularea de 77 GZ (Hsieh, Roberts, Armbrust, 1974). Insecta adultă rămâne în câmpurile de lucernă timp de 1-3 săptămâni pentru hrănirea suplimentară sau până la prima recoltare, apoi migrează spre locurile potrivite pentru estivare, de obicei, în afara câmpurilor de lucernă (Tysowsky, Dorsey, 1970; Kuhar, Youngman, Laub, 2010;Zahiri et al., 2014) unde are loc dezvoltarea sexuală, iar activitatea metabolică este încetinită (Litsinger, Apple, 1973). Datorită schimbării fotoperioadei, insectele adulte devin din nou active, migrează înapoi spre câmpurile de lucernă, ating maturitatea sexuală și încep depunerea pontei (Kuhar, Youngman, Laub, 2000).Protapion apricans.
