Antagonistic actions of juvenile hormone and 20-hydroxyecdysone within the ring gland determine developmental transitions in Drosophila

Abstract: SignificanceIn vertebrates, steroid hormones regulate developmental transition from juveniles to adults. Insect steroid hormone, 20-hydroxyecdysone (20E), coordinates with juvenile hormone (JH) to regulate metamorphosis; however, the precise cross-talk mechanism is not well understood. Here, we report that JH and 20E antagonize each other’s biosynthesis in a major endocrine organ of Drosophila larvae: JH suppresses ecdysone biosynthesis and inhibits metamorphosis, whereas 20E suppresses JH biosynthesis and pro… Show more

“…Recent reports indicate that JH can inhibit ecdysone production in holometabolan insects, such as the fly D. melanogaster and the silkmoth Bombyx mori. The effect is mediated by Kr-h1, which represses the expression of ecdysteroidogenic genes (37,38). Therefore, it is plausible that the reduced expression of nvd, phm, and dib in dsMyo-treated cockroaches could be due to an increase in Kr-h1 expression resulting from Myo depletion ( Fig.…”

“…This supports the hypothesis that FTZ-F1 helps enhance phm and dib transcription in the PG of B. germanica. Finally, decreased expression of ecdysteroidogenic genes in Myodepleted insects might be mediated by the increased expression of jhamt and Kr-h1 because JH and Kr-h1 inhibit the expression of these genes (37,38). We explored this possibility through experiments examining Myo in N6.…”

“…Hence, JH suppresses ecdysone biosynthesis and inhibits metamorphosis, whereas 20E suppresses JH biosynthesis and promotes metamorphosis. Antagonistic actions of JH and 20E at the hormone‐biosynthesis levels determine developmental transitions in D. melanogaster (Liu et al ., ; Zhang et al ., ).…”

“…Thus, JH prevents 20E‐induced metamorphosis and is therefore referred to as the status quo hormone. During middle and late juvenile (larval or nymphal) stages, JH coordinates with 20E to maintain the juvenile status by preventing metamorphosis; in the final juvenile instar, insects undergo pupation or adult morphogenesis because of a sharp decrease in JH titer (Jindra et al ., ; Belles & Santos, ; Jindra et al ., ; Liu et al ., ). In adults, JH participates in regulating reproduction, including previtellogenic development, vitellogenesis and oogenesis (Roy et al ., ).…”

“…In the presence of JH, 20E cannot trigger pupation but ecdysis to another larva Liu et al, 2018). In the absence of JH, 20E initiates the pupal programme, in which larval structures are transformed into imaginal ones and new pupal cuticle is deposited.…”
