Fourteen strains of Trichoderma spp. were isolated from Algerian desert soils and assessed for their antagonistic activity against Fusarium crown and root rot of wheat. Biocontrol efficiency of Trichoderma spp. was studied by in vitro and in vivo based bioassay against three pathogenic species: F. culmorum, F. graminearum and F. verticillioides. In vitro based bioassay (dual culture) results obtained with all Trichoderma spp. isolates showed significant decrease in colony diameter of Fusarium species compared to the control. The highest percentages of reduction in colony diameter were obtained with T. harzianum Thr.4 causing a growth reduction of 70.68%, 67.05 and 70.57% against F. culmorum, F. graminearum and F. verticillioides, respectively. All Trichoderma spp. isolates were able to overgrow and sporulate above F. culmorum colonies but no overgrowth was observed with F. graminearum and F. verticilliodes. The seed treatment by Trichoderma spp. isolates before sowing in a soil already infested by the pathogens led to a significant decrease of disease severity compared to the untreated control. The highest disease index decrease (>70%) was obtained with two isolates of T. harzianum (Thr.4 and Thr.10) and T. viride Tv.6 against the three fungal pathogens. Lytic enzymes production by Trichoderma spp. isolates was tested in liquid cultures containing fungal cell walls of each pathogen as sole carbon source. Higher levels of protease and chitinase activities were induced by hyphal cell walls of F. graminearum than cell walls of F. verticillioides and F. culmorum. T. harzianum Thr.4 exhibited the highest enzyme activities with hyphal cell walls of F. graminearum and F. culmorum. However, in the medium amended with cell wall of F. verticillioides, maximal lytic activities were recorded for T. viride Tv.6.

The aim of this study was to isolate and to identify the pathogenic fungi accompanying interior in hard wheat seeds, and to evaluate the antagonism capability in vitro of (Trichoderma harzianum) against those pathogenic isolated fungi. Three samples of local hard wheat seeds (Triticum durum Desf) were brought from Oum-elbouaghi (Algeria) silos yields, follower for varieties: Vitron, Waha, and Cirta. ).They were stored for one year. The results of isolation on potato-dextroseagar (PDA) medium and after identification of fungal accompanying interior in the seeds clarified about (83) fungus isolates following for species next : Fusarium acuminatum (1 isolate) in a ratio of (1.2%), and Alternaria(82 isolates) with a ratio of (98.795 %).The Alternaria isolates apportioned among two species: Alternaria alternata with 39 isolates(12 isolates for: Alternaria alternata1 and 27 isolates for : Alternaria alternata2) and Alternaria infectoria with 43 isolates).And, besides it was clearly seen that the Cirta variety was a lowest infected with fungi(19 isolates) in ratio of (22.89 %), and both varieties Waha and Vitron with(32 isolates)for each one in a ration equal (38.554 %) . The results of the antagonistic capability in vitro of (Trichoderma harzianum) against isolated fungi, showed that: 1-The direct confrontation in vitro of Trichoderma harzianum against the different fungus isolates on PDA medium, showed that: aIn the fourth day of the experiment, Trichoderma harzianum inhibited the mycelia growth of different pathogenic fungi in dual cultures, with different ratios, it was equal to: 6.66% for Fusarium acuminatum, and 23%, 26.66% and 40% for Alternaria alternata1, Alternaria alternata2, and Alternaria infectoria, respectively. b-The microscopic observations of the different interactions hyphal between Trichoderma harzianum and different fungus isolates, showed that the antagonistic fungus affected on Alternaria species with the decomposition phenomenon (Lyses), where it analyzed the mycelia and spores of Alternaria altrenata2 and Alternaria infectoria, while it analyzed the mycelia of Alternaria alternata1, compared with control. 2-The remote confrontation in vitro of Trichoderma harzianum against the different fungus isolates on PDA medium showed that : a-The volatile metabolic substances of Trichoderma harzianum affected the growth of the different pathogenic fungi, with different ratios over the seven days of treatment, compared with control.b-The microscopic observations Noted that the volatile metabolic substances of Trichoderma harzianum affected some isolated fungi with mycelia analysis prevent the spore's formation of the Alternaria alternata2, while stopped the spore formation of Fusarium acuminatum, compared with control.

The aim of this study is to clarify the antagonism capability in vitro of the antagonistic fungus (Trichoderma harzianum) against the pathogenic fungus, four isolates of pathogenic fungus associated internally with the solid wheat seeds ( Triticum durum )Desf , follower for species: Botrytis cinerea, Cladosporium sp, Stemphylium botryosum and Alternaria sp, were brought. The antagonistic sample (Trichoderma harzianum) was isolated from the wheat plant soil. The results showed that:The direct confrontation of Trichoderma harzianum against the different fungus isolates in vitro on PDA medium, showed in the third day of the experiment an inhibition in the pathogenic mycelia growth, with a different ratios, it was equal to: 41.66% and 50% for Stemphylium botryosum and Cladosporium sp, respectively, and amounted in the fourth day to 56.52% and 57.14% , for Botrytis cinerea and Alternaria sp, respectively, did not show any growth of the different pathogenic fungi when re-planting a disk from the interaction hyphal areas between the antagonistic fungus and the pathogenic fungus from the different dual cultures, while the antagonistic fungus was grown. The microscopic observations of the different interactions hyphal showed that the antagonistic fungus was affected on the pathogenic fungi with a several biological forms: Decomposition phenomenon (Lyses): the antagonistic fungus was analyzed the mycelia and spores of Cladosporium sp, while was analyzed the mycelia and damped the spore formation in the other pathogenic fungi, compared with control. Parasitism phenomenon (Mycoparasitism): it was found that the hyphae of Trichoderma harzianum has formed Haustoria on the cell walls of Stemphylium botryosum hyphae and they penetrated within them. The remote confrontation showed that the volatile metabolic substances of the antagonistic fungus affected the growth of the pathogenic fungi , with a different rates over the seven days of treatment, it peaked after two days of treatment to reach 13.33% and 50% in Botrytis cinirea and Cladosporium sp, respectively, and decreased to 08.33% in the fourth day in Botrytis cinirea and scored in the fifth day a ratio equal to 25.42% for Cladosporium sp, and was scored the maximum ratio in the third day in Stemphylium botryosum to 23.07%,and lowered to 07.93% in the seventh day, but in the Alternararia sp has recorded the lowest inhibition percentage to 05.55% in the third day and 05.76% in the seventh day. The microscopic observations Noted that the volatile metabolic substances of the antagonistic fungus was affected the pathogenic fungi with a several modes, their were with the mycelia analysis and prevent the spore formation in the Alternararia sp, while was analyzed and aggregated the spores in the Cladosporium sp, and stopped only the spore formation in both Botrytis cinirea and Stemphylium botryosum, compared with control.
