Anomalous bone changes in ovariectomized type 2 diabetic rats: inappropriately low bone turnover with bone loss in an estrogen-deficient condition

Abstract: Estrogen deprivation accelerates bone resorption, leading to imbalance of bone remodeling and osteoporosis in postmenopausal women. In the elderly, type 2 diabetes mellitus (T2DM) frequently coexists as an independent factor of bone loss. However, little is known about the skeletal changes in a combined condition of estrogen deficiency and T2DM. Herein, we performed ovariectomy (OVX) in nonobese Goto-Kakizaki (GK) T2DM rats to examine changes associated with calcium and phosphate metabolism and bone microstruc… Show more

“…In addition, various new genes were determined as nanohydroxyapatite target genes including insulin-like growth factor I and II (IGF-I and IGF-II) [60]. Aeimlapa et al [90] commented that IGF-I, which belongs to prominent cytokines for bone formation, is significantly increased in OVX rats. IGF-I induces alkaline phosphatase activity in primary rat calvaria cell culture, and IGF-II elevates alkaline phosphatase activity in human osteoblasts cultures [91,92].…”

“…При сочетании менопаузы и СД скорость костного обмена имеет свои особенности в силу разнонаправленного влияния дефицита эстрогенов при менопаузе и сложного патогенеза СД. Многие экспериментальные исследования указывают на замедление костного обмена при СД и менопаузе [28][29][30]. Влияние сахароснижающих препаратов на костную ткань в экспериментальных исследованиях довольно широко обсуждается в литературе.…”

“…Bone microstructural defect in 4-month-old male BKO vs. WT mice was confirmed by bone histomorphometry, using OsteoMeasure system (version 4.10; OsteoMetrics, Atlanta, GA, USA) equipped with Eclipse Ni-U light/fluorescent microscope (Nikon), as previously described 16 , 34 . Trabecular bone microstructural parameters consisted of trabecular bone volume normalized by tissue volume (bone volume fraction, BV/TV; %), trabecular number (Tb.N; mm −1 ), trabecular separation (Tb.Sp; µm) and trabecular thickness (Tb.Th; µm).…”
