Angiotensin II type 2 receptors mediate inhibition of mitogen-activated protein kinase cascade and functional activation of SHP-1 tyrosine phosphatase

Abstract: Angiotensin II type 2 (AT2) receptors are involved in the inhibition of cell proliferation as well as in apoptosis and neuronal differentiation, through intracellular signalling pathways that remain poorly defined. The present study examines the effect of AT2-receptor stimulation on growth-factor-induced pathways leading to the activation of mitogen-activated protein (MAP) kinases. In N1E-115 neuroblastoma cells, AT2 receptors inhibit the activity of MAP kinases induced by serum as well as by epidermal growth … Show more

“…Consistent to the previously reported observation (8), stimulation of AT 2 receptor by 0.1 M of Ang II in COS-7 cells cotransfected with HA-AT 2 and SHP-1 induced rapid SHP-1 activation as assayed in immunocomplexes prepared with anti-SHP-1 antibodies (Fig. 1A).…”

“…However, the AT 2 -mediated SHP-1 activation is PTX-insensitive (8). These contradictory findings do not support an involvement of G ␣i proteins in AT 2 -mediated SHP-1 activation.…”

“…In quiescent N1E-115 neuroblastoma cells expressing only the AT 2 receptor, basal MAPK activity was not affected by Ang II treatment. However, Ang II prevented the fetal calf serum-as well as epidermal growth factor-induced activation of ERK1 and ERK2 (Bedecs et al 1997). Furthermore, Ang II-induced neurite outgrowth is associated with changes in the levels of microtubuleassociated proteins (MAPs), such as up-regulation of MAP2c in NG108-15 cells and MAP2 in PC12 W cells, and downregulation of MAP1B in PC12 W cells (Laflamme et al 1996;Stroth et al 1998).…”

“…Different exposure times are shown in order to detect Cdk4 in all the conditions tested. Immunoblots were quantified by densitometric analysis and the mean half-lives were calculated from three different experiments Huang et al, 1996;Yamada et al, 1996;Bedecs et al, 1997), and in the heart of transgenic mice overexpressing the AT 2 receptor (Masaki et al, 1998). However, it is unlikely that inhibition of the ERK mitogen-activated protein kinase pathway is responsible for the antiproliferative action of the AT 2 receptor, at least in rat fibroblasts.…”
