Angiotensin II attenuates renal cortical cyclooxygenase-2 expression

Abstract: We have previously shown that in rat renal cortex, cyclooxygenase-2 (COX-2) expression is localized to cTALH cells in the region of the macula densa, and that dietary salt restriction increases COX-2 expression. Administration of the angiotensin converting inhibitor, captopril, further increased COX-2 mRNA and renal cortical COX-2 immunoreactivity, with the most pronounced expression in the macula densa. Administration of an AT1 receptor antagonist, losartan, also significantly increased cortical COX-2 mRNA ex… Show more

“…Our data now show that in the absence of calcineurin inhibitors low-salt diet and a combination of low-salt diet with an ACE inhibitor increase renin secretion as well as renin-and COX-2 mRNA expression in parallel, which is in accordance with previous studies (7,8,25). In the presence of CsA, low-salt diet and the combination of low-salt diet and ACE inhibitor stimulates renin secretion and renin expression, whereas Figure 8.…”

“…The data obtained in this study clearly demonstrate that calcineurin inhibitors not only decrease basal COX-2 gene expression in the renal cortex and medulla of rats but also attenuate the well-established noninflammatory induction of renocortical COX-2 expression, particularly in the TALH/macula densa structures, by low-salt diet or AngII antagonists (2,7,8). Inhibition of COX-2 gene expression was associated by a decline of COX-2 protein expression as well as by a fall of tissue PGE 2 levels.…”

“…This phenomenon is probably a feedback response to the low renin, because decreased renin and angiotensin II levels should lead to decreased angiotensin II-mediated inhibition of COX-2 (36). What, then, is causing the renin to be low?…”

“…12 In addition, an attenuation of COX-2 expression in rat renal cortex by Ang II was observed. 13,14 Worth noting, in aorta from spontaneously hypertensive rats, an animal model of oxidative stress, increased ROS production was shown to activate COX-1 leading to the production of endoperoxides, which, in turn, stimulate TP receptors on vascular smooth muscle cells. 8 Based on these findings, ROS might be hypothesized as a mechanism whereby Ang II interacts with the COX pathway to induce endothelial dysfunction.…”
