The effects of salmon angiotensin I (sAI) and human angiotensin II (hAII) on in vitro ovulation of preovulatory (preOV) brook trout (Salvelinus fontinalis) follicles were investigated. Both angiotensins increased levels of ovulation above that in controls after 12 hours of incubation. The increase was statistically significant in incubates with greater than 1 μM hAII. The effects of the angiotensins on follicle contraction were also studied indirectly by measuring the decrease in weight of punctured follicles taken prior to germinal vesicle breakdown. At 1 μM, both angiotensins significantly decreased the weight of punctured follicles after 16 hours of incubation. The angiotensin-stimulated decrease in weight was not blocked by indomethacin (10 μg/ml), indicating that follicle contraction was not prostaglandin-dependent. The data indicate that angiotensins might be directly involved in brook trout ovulation and the stimulatory effects of angiotensins on ovulation may be attributed to their effects on follicle contraction.

The corticotropin-releasing factor (CRF), arginine-vasopressin (AVP), atrial natriuretic peptide (ANP), and Angiotensin II (Ang II) interact, regulating the central sympathetic system, the hypothalamic-pituitary-adrenal (HPA) axis, and the central behavioral mechanisms involved in the response to stress and the development of mood disorders. Non-peptidic, orally active AVP V 1 , V 3 , or CRF 1 receptor antagonists reduce anxiety in animals. ANP is anxiolytic when administered to rodents and humans with anxiety disorders. These compounds are being developed for the therapy of anxiety and depression, but their clinical efficacy has not yet been established.Brain Ang II, a physiological ANP antagonist, promotes CRF and AVP release and stimulates the peripheral and central sympathetic systems. The degree of Ang II AT 1 receptor stimulation determines the response to stress. An orally active, non-peptidic Ang II AT 1 receptor antagonist blocks CRF and AVP release during stress, prevents the sympathoadrenal and hormonal stress reaction, the cortical CRF 1 and benzodiazepine receptor alterations, facilitates the effects of ANP, prevents stress-induced gastric ulcerations, and is anxiolytic in rodents. CRF, AVP, ANP, and the sympathetic system are intimately linked parts of a fundamental regulatory mechanism controlled by the brain Ang II system through AT 1 receptor stimulation, and should be studied together. Selective, safe antagonists of Ang II AT 1 receptors, with central effects after oral administration, have been actively used for the treatment of hypertension and should be considered also as potential anti-stress, anti-anxiety, and antidepressant compounds with properties similar or superior to the CRF and AVP antagonists or the ANP agonists under development. RESUMOAlém das gonadotrofinas e esteróides gonadais, outros fatores participam da regulação da função ovariana; entre estes incluem-se fatores de crescimento e diversos peptídeos regulatórios, como os peptídeos natriuréticos e o sistema renina-angiotensina (SRA), Além do conhecido SRA sistêmico, vários órgãos, entre eles os ovários, contêm SRA locais. Todos os componentes do SRA foram descritos em ovários: angiotensinogênio, angiotensina-I, angiotensina-II (Ang-II), renina, enzima conversora de angiotensina (ECA) e receptores AT 1 e AT 2 , A Ang-ll estimula a ovulação, maturação dos oócitos e produção de estradiol, progesterona e prostaglandinas; a produção de Ang-ll, por sua vez, é estimulada pela hCG. Entretanto, a inibição da produção de Ang-ll por inibidor de ECA não altera a esteroidogênese ou a ovulação. Tais efeitos poderiam ser devidos à angiotensina-(1-7), um peptídeo do SRA cuja formação pode ocorrer independente da ECA. Resultados preliminares mostram que a angiotensina-(1-7) estimula a produção de estradiol in vitro. Finalmente, há evidências da participação do SRA em disfunções ovarianas, tais como a síndrome de hiperestimulação ovariana (SHO) e a síndrome dos ovários policísticos (SOP). Na SHO foi demonstrado que o tratamento com gonadotrofinas estimula o SRA ovariano e que o uso de inibidores da ECA diminui a incidência e a gravidade da síndrome. Na SOP foi demonstrado aumento de renina e Ang-ll ovarianas, enquanto o aumento da pró-renina plasmática se correlaciona com a concentração de andrógeno, uma característica básica da síndrome. Assim, o SRA parece ser um sistema importante na fisiologia e fisiopatologia ovariana. ABSTRACTGonadotropins and gonadal steroids play essential roles in the ovarian function. Several factors, including growth factors and regulatory peptides, have also been involved in this function. Recently, natriuretic peptides and the renin-angiotensin system (RAS) have been shown to participate in this process. Local RAS has been described in several organs including the ovaries. All the components of the RAS have been identified in the ovaries: angiotensinogen, angiotensin-I (Ang-I), angiotensin-II (Ang-II), renin, angiotensin-converting enzyme (ACE), and AT 1 and AT 2 receptors. Ang-II stimulates ovulation, maturation of oocytes, and production of estradiol, progesterone and prostaglandins. HCG stimulates the ovarian production of Ang-II. The inhibition of Ang-II production by ACE inhibition, however, does not change steroidogenesis and ovulation. Such effects could be due to Angiotensin-(1-7), a recently described RAS peptide, which can be formed by an ACE independent pathway. Preliminary results show that Ang-(1-7) induces estradiol release in rat ovary perfused in vitro.
